🚀 Discovering a New Stable Form with Crystal16 🌟 Excited to share a recent publication in the Journal Organic Process Research & Development, titled " Characterization, Solubility, and Hygroscopicity of BMS-817399", featuring the use of Crystal16 multi-reactor system. This study identified and characterised a new, stable anhydrous form (Form 2) of BMS-817399, demonstrating superior stability compared to the original monohydrate (Form 1). Crystal16 was used to determine the solubilities of BMS-817399 in ethanol, isopropanol, acetone, and acetonitrile by heating slurries of the compound at a rate of 0.3°C/min while monitoring solution transmissivity. Recrystallisation was observed only in acetonitrile, leading to the identification of a new, stable anhydrous form (Form 2). 🔬 Key Highlights: Discovery of a New Stable Form: A new, physically stable anhydrous form (Form 2) of BMS-817399 was identified, which is more reliable for pharmaceutical use compared to the original monohydrate (Form 1). Precise Solubility Measurements: Using the Crystal16, solubilities of Form 1 in various solvents (ethanol, isopropanol, acetone, and acetonitrile) were accurately determined. Recrystallisation occurred only in acetonitrile, confirming the existence of Form 2. Thermodynamic Studies: The van’t Hoff equation was applied to solubility data obtained from Crystal16, to understand the solution thermodynamics, revealing that Form 2 has favorable solute-solvent interactions. DSC and TGA confirmed Form 1 as a monohydrate and Form 2 as anhydrous, highlighting structural differences and stability. Dynamic vapor sorption studies showed Form 1's water content varies with humidity, while Form 2 is more stable.
