Unlike classic experiment design approaches, on-the-fly optimal experimental design increases the amount of information derived from process runs by exploring the temporal domain. Running a high-throughput bioreactor platform is challenging, but running it for a perfusion mammalian cell culture process is even more difficult. Using a digital twin to support the planning and operation of the runs increases the insight into the process evolution and the information derived from each reactor run, as shown in the latest publication "Self-Driving Development of Perfusion Processes for Monoclonal Antibody Production". The core component of the digital twin is the predictive hybrid model that renders what-if analysis, as well as the exploration of meaningful unseen process regions. In the biopharmaceutical industry, accurately predicting critical quality attributes (CQAs) while minimizing costly experimental runs has been a long-standing challenge. Traditional “black box” models are the industry standard but are still unable to completely understand the complexities of bioprocesses. Together with Bristol Myers Squibb, we evaluated experimental data from a complex upstream mAbs bioprocess using DataHow’s AI-enabled hybrid process models. The results show that hybrid (AI) models offer significantly more insight than black box models: • 22% increase in prediction accuracy across CQAs • 3x fewer experiments required to predict CQAs accurately • Richer process knowledge, insights, and control compared to traditional models Want to learn more about how hybrid models can impact process development? Reusing existing process data to streamline new process development activities can significantly reduce the experimental burden. In our latest work, we show how transfer learning and meta-learning applied to historical process data can transform how we design and predict new bioprocesses. By leveraging knowledge transfer models, we introduce a novel method, calibration design, that optimizes process design for new campaigns. Our results suggest that: • Process understanding is significantly improved when horizontal knowledge transfer methods are used in lieu of single process models; • Calibration designs allows to systematically carry knowledge over to new process development campaigns; • Gaussian Process model performance is better when using Latin Hypercube Sampling for data generation than Definite Screening Designs.
