📍 POSTER | AI-enabled hit selection on human pancreatic #cancer #organoids grown in #Gri3D 🔬 Measuring the impact of anticancer compounds and their combinations relies heavily on robust ex vivo assays. To this end, patient-derived organoids (#PDOs) are increasingly utilized as effective models for ex vivo testing. In this collaborative work with Molecular Devices and Core Life Analytics, the #Gri3D platform was used to perform #drug #screening on pancreatic #PDOs. Its hydrogel-based microcavities enable the formation of uniform microtissues, all aligned within the same focal plane, making it an ideal choice for high-throughput #screening. Human #pancreatic cancer PDOs were exposed to various anticancer compounds at different doses, and their responses were monitored using the #ImageXpress® system for high-content confocal imaging. 📊 The organoid responses were analyzed with #INCarta® software, and the phenotypic features related to each organoid were further processed with #StratoMineR® software, allowing for robust and unbiased data analysis. 🌍 | Last week in rainy #London was a memorable one for SUN bioscience! 🌧️ 🇬🇧 🤝 Michelle Ballabio had the pleasure of attending both the Molecular Devices Users Meeting and the ELRIG UK Drug Discovery 2024 meeting. It was an incredible opportunity to connect with collaborators and #Gri3D users and to listen to some truly inspiring talks. ✨ A key highlight was hearing our clients share their successes using the #Gri3D platform: • Allan Sauvat, researcher at the Gustave Roussy Institute, introduced their research using #Gri3D to study the automated 3D kinetic profiling of dendritic cells in the tumor microenvironment, featuring some stunning images 🔬 • Angeline Lim Lim, from Molecular Devices Devices highlighted #Gri3D for AI-based screening of pancreatic cancer organoids 🤖. • The team of Luigi Aloia at AstraZeneca, represented by Lambert Montava Garriga, PhD, showcased #Gri3D in their poster at #ELRIGDD24 on an innovative workflow to integrate a multidisciplinary CRISPR organoid platform for target and #DrugDiscovery in oncology🧬. 📚 PUBLICATION | Retinal #organoids grown for 26 days in 1600µm microwells #Gri3D plates to obtain 7 microtissues per well. Many genetic mutations cause dysfunction and death of rod and cone #photoreceptors (PR), the two key photosensitive cell types in the retina, leading to vision decline and often blindness. While many efforts are made to unravel mechanisms behind PR loss, no therapy exists to robustly protect photoreceptors, highlighting the need to develop improved in vitro #retinal models. Towards that goal, scientists from EPFL and University of Lausanne - UNIL reported a tissue-engineering approach to accelerate and standardize the production of #retinalorganoids by culturing mouse embryonic stem cells in microcavities – yes, that’s one of the first publication describing #Gri3D.
