Cytokine profile of anti-spike CD4+ T cells predicts humoral and CD8+ T cell responses after anti-SARS-CoV-2 mRNA vaccination. Researchers found that a cytokine signature composed of four cytokines (IL-2, TNF-α, IP10, IL-9) excluding IFN-γ could effectively predict the CD8+ T cell response. They also identified a correlation between CD4+ T lymphocyte-produced IL-21 and the humoral response. A new improved way to identify immunogenic neoantigens! The VACINUS in silico neoantigen prediction workflows incorporating pMHC binding affinity and TCR reactivity to pMHC, to identify neoantigens better at activating an immune reaction against cancer cells, leading to more effective cancer vaccines. The study validated these workflows in patients with hepatocellular carcinoma and a mouse melanoma model, confirming their predictive abilities. The results demonstrated that vaccination with the predicted neoepitopes induced neoantigen-specific T cell responses, leading to tumor suppression. #Dextramer® reagents were used to isolate neoantigen-specific CD8+ T cells for further analysis of T cell subtypes and states contributing to the immune responses using scRNA-seq and TCR-seq. The initial SARS-CoV-2-specific CD8+ T cell response could predict the development of long-term complications from SARS-CoV-2 infection, such as long COVID. This study investigated SARS-CoV-2-specific CD8+ T cell responses in COVID-19 patients with severe disease and vaccinated healthcare workers. MHC #Dextramer® reagents were used to analyze the CD8+ T cell responses against 26 SARS-CoV-2-derived epitopes. The total number of SARS-CoV-2-specific CD8+ T cells was associated with protection against long COVID. Antigen-specific T cells induce and track Alzheimer’s-like neurodegeneration! This study recently published in PNAS indicates that age-related T cells generate neurodegeneration with characteristics of Alzheimer’s disease (AD) in mice, with distinct T cell functions required for the pathological initiation and neurodegenerative progression. Additionally, the researchers found that the analogous age-related, antigen-specific CD8+ T cell population was significantly elevated in the brains of human AD patients, and its loss from blood corresponded better to sporadic AD and cognitive decline than the promising biomarker candidate plasma pTau-217. This study identifies an age-related T cell-mediated factor acting upstream of plaque formation/tau to initiate AD-like pathophysiology, with implications for earlier diagnosis and more effective treatment of the disease. #Dextramer® reagents were used to quantify antigen-specific T cells by flow cytometry. Congratulations Adaptimmune on this major milestone!🎉 TECELRA® (afamitresgene autoleucel, or afami-cel) has been approved for advanced MAGE-A4+synovial sarcoma in adults with certain HLA types who have received prior chemotherapy. Afami-cel works by collecting and engineering a patient’s T cells to target the MAGE-A4 protein on cancer cells, then reinfusing them to selectively destroy cancer cells while sparing normal cells. Colon cancer derived antigen-specific T cells have an anti-metastatic effect! This research published in Science Immunology used metastatic colorectal cancer preclinical models to show that orthotopically implanted primary colon tumors exert a colon-specific antimetastatic effect on distant hepatic lesions. Enterotropic α4β7 integrin–expressing neoantigen-specific CD8 T cells were key components of the antimetastatic effect. #Dextramer® reagents were used to stain CD8+ T cells specific for an MC38-neoantigen and investigate the proportion of these antigen-specific T cells in the tumor as well as peripheral compartments including the blood, spleen, and lymph nodes. First-in-Human dose escalation trial of Anti-MAGEA1 TCR-T Cell Therapy in Advanced Solid Tumours! This trial evaluated the clinical safety and efficacy of IMA202, which consists of autologous CD8+ T cells genetically modified to express a MAGEA1-specific TCR. #Dextramer® reagents were used to quantify the MAGEA1- specific T cells for dose determination. Overall, the trial demonstrated a manageable safety profile, and no dose-limiting toxicities were observed. There was evidence of biological and potential clinical activity of the engineered TCR-T cells targeting the cancer testis antigen MAGEA1, which is expressed in several solid tumor indications. IMA202 T cells persisted in peripheral blood for several weeks to months and were detected in tumor tissue.
