    Ever wondered how close we are to detecting the recurrence of high-risk, early-stage cancers? Before they become an issue? Well, our team recently attended the reputable Update '24 Symposium organized by MKOT. So, we might be able to shed light on that… Now, ctDNA-testing is already being used to evaluate the effectiveness of certain cancer therapies. For example, the MonarchE trial compared the effectiveness of combined abemaciclib - standard hormone therapy with hormone therapy alone... ...by checking the blood-levels of patients’ ctDNA (as detailed in Professor Sherene Loi’s presentation). But the potential of ctDNA testing goes beyond determining the effectiveness of various treatments. While more research is needed the use of ctDNA testing may also be able to diagnose the return of cancer earlier in high-risk breast cancer patients after surgery. In fact, ctDNA has even been identified as a key biomarker across various tumor types – not “just” breast cancer. This means it can have both a predictive and a prognostic value across a range of tumors – potentially leading to more precise & effective personalized cancer treatments and diagnostics in the future.     Most current cancer treatments have 3 major issues… 1) Non-specific targeting ↳ Damage to healthy tissues leads to side-effects & reduced effectiveness. 2) Low effectiveness ↳ Tumors can develop genetic resistance, leading to proliferation, recurrence, or progression. 3) Nasty side-effects ↳ Decreases the quality of life for patients, while limiting the dose & duration of treatments. But what if we can overcome these challenges with novel therapeutic targets? Well, our team is making significant progress in the field of neoantigen research to do just that! Neoantigens? Yes! These are unique markers on cancer cells, absent from normal tissues. They arise due to mutations in tumor cells – posing as highly specific potential targets for both the immune system and novel therapies. This specificity may lead to treatments that: • Are highly personalized ↳ to the individual’s specific tumor profile. • Have better effectiveness ↳due to targeting tumors specifically. • Come with reduced risks ↳ of off-target effects or collateral damage. To make this possible, we’re currently in the process of: • Identifying specific neoantigens that could be potential therapeutic targets. • Developing methods to identify potent neoantigens from FFPE samples.     Can therapies targeting bacteria improve cancer outlooks? Fusobacterium Nucleatum is a bacteria often found in the human gut & oral cavity. But unfortunately – according to a recent review published in Cancer Biology & Therapy by the Taylor & Francis Group – it may contribute to the development, progression, and treatment resistance of various cancers. So, what’s the good news? According to the same study, reducing F. Nucleatum colonization has shown immense promise in slowing tumor growth and improving survival rates in animal studies. Now, this can be done in several ways, such as: • Targeted antibiotic therapy to eliminate F. Nucleatum • Fecal microbiota transplantation to improve gut barrier integrity • Personalized probiotics & prebiotics to decrease F. Nucleatum colonization What’s more, the presence of F. Nucleatum also shows promise as a potential biomarker for early cancer detection and/or cancer risk assessment. This means that advanced NGS-based microbiome diagnostics (such as ours) can potentially become a convenient, accurate, and comfortable tool to screen for a variety of cancers very soon.     What’s the future of breast cancer monitoring? That’s what our Chief Scientific Officer, Katalin Priskin talked about when she took the stage at the “2. Liquid Biopsy Seminar” organized by Clinomics Europe. Kati introduced BC-Monitor – our custom-developed breast cancer monitoring solution based on liquid biopsy and next-generation sequencing. From a simple blood sample, BC-Monitor analyzes circulating tumor DNA, providing deep insights into the molecular changes of cancer. Ultimately, this helps oncologists to: • Identify the recurrence or metastasis of breast cancer earlier • Monitor the effectiveness of a given breast cancer therapy • Indicate whether it’s time to change to a new therapy • Choose the right therapy for each individual patient As a minimally invasive approach, BC-Monitor allows for more frequent (and more comfortable) monitoring of breast cancers compared to traditional biopsies and imaging techniques. Moreover, BC-Monitor offers an accurate real-time picture of pathological processes at a molecular level – signalling mutations & changes well before clinical progression is evident.     Can childhood asthma originate… from the gut? Childhood asthma is one of the most common chronic illnesses in early life – affecting approximately 300 million children worldwide. The most common symptoms are shortness of breath, wheezing, and coughing. So, naturally, the primary focus of asthma research for many years has been focused on the lungs. But according to a scientific study published in Science Translational Medicine, when it comes to the causes (and possible treatments) of asthma, there may be another area worth exploring... The gut! In the study, researchers found a strong link between early-life gut dysbiosis and the development of childhood asthma. More precisely, they found that a lower percentage of certain strains of gut bacteria – namely: Lachnospira, Veillonella, Faecalibacterium, and Rothia – in the first few months of a child’s life had a high predictive value to the development of asthmatic symptoms a few years later (possibly due to lower levels of intestinal acetate-production). Could this also imply that gut bacteria might play a crucial role in the effective treatment of childhood asthma? While more research is needed to give a definite answer to this question… …discoveries like these highlight the diverse effects our gut bacteria can have on our health & well-being – and the importance of obtaining accurate information about our gut microbiomes.
