🚀Exciting developments from EHA 2024! Dr Hanne Norseth from the Oslo Myeloma Center presented the results of the phase I/II TG01 study. This study explores the safety and efficacy of the TG01/QS-21 vaccine in patients with KRAS or NRAS mutations. While the vaccine was well-tolerated, further evaluation is needed to determine its clinical efficacy. Stay tuned for more updates on this innovative approach in multiple myeloma treatment. Happy to announce the publication of my latest article, "Nanotechnology strategies to address challenges in topical and cellular delivery of siRNAs in skin disease therapy" in the esteemed journal "Advanced Drug Delivery Reviews" by Elsevier as part of my doctoral work. I thank Maria Vitoria Bentley for the supervision and support and the co-authors, Milena Finazzi Morais, for their dedication and commitment. In this article we bring together the main challenges of topical and cellular delivery of siRNAs, explore strategies to overcome them, and finally, an overview of the application and therapeutic effects of siRNAs (delivered in nanoparticles and physical methods) in wound healing diseases, skin cancer, psoriasis, dermatitis among other dermatological diseases is shown. We hope that this review will expand knowledge about the delivery barriers of siRNA (and other nucleic acids) and provide new insights to improve their bioavailability and therapeutic effects, and thus bring new hope for therapy of dermatological diseases. Nasal tau immunotherapy clears intracellular tau pathology and improves cognitive functions in aged tauopathy mice Sagar Gaikwad et al. SCIENCE TRANSLATIONAL MEDICINE 3 Jul 2024 Vol 16, Issue 754 Gli aggregati tau patologici causano il declino cognitivo nelle tauopatie neurodegenerative, inclusa la malattia di Alzheimer (AD). Questi aggregati sono prevalenti all'interno dei compartimenti intracellulari. Le attuali immunoterapie tau hanno mostrato un'efficacia limitata nell'eliminazione degli aggregati tau intracellulari e nel miglioramento delle funzioni cognitive negli studi clinici. In questo studio è riportato l’effetto di un anticorpo monoclonale tossico specifico per la conformazione tau-2 (TTCM2), che ha riconosciuto selettivamente gli aggregati tau patologici nei tessuti cerebrali di pazienti con AD, demenza con corpi di Lewy (DLB) e paralisi sopranucleare progressiva (PSP). TTCM2 ha inibito in maniera significativa l'attività di semina tau, un meccanismo essenziale alla base della progressione della tauopatia. Per colpire efficacemente gli aggregati tau intracellulari e garantire un rapido rilascio al cervello, TTCM2 è stato caricato in micelle (TTCM2-ms) e somministrato per via intranasale. TTCM2-ms somministrato per via intranasale entra in modo efficiente nel cervello nei topi hTau-tauopatia, prendendo di mira la tau patologica nei compartimenti intracellulari. Inoltre, una singola dose intranasale di TTCM2-ms ha eliminato efficacemente la tau patologica, aumentando le proteine sinaptiche e migliorando le funzioni cognitive nei topi anziani con tauopatia. TRIM21 è risultato essenziale per la clearance mediata da TTCM2-ms della patologia tau. Questo studio potrebbe essere prezioso nella progettazione di immunoterapie tau efficaci per l’AD e altre tauopatie. https://lnkd.in/d43W2x-M

Excerpt from PD03-02: #QS11 is a small molecule #ARFGAP1 inhibitor and activator of #Wnt/ß-catenin signaling pathway, sourced from Tocris Bioscience, promoting #prostate smooth muscle contraction. These interesting data may help to explain our findings regarding immunomodulatory imde drugs (IMiDs), presented at this year‘s #EAU24 in #Paris. #IMiDs interfere with the Wnt/ß-catenin signaling pathway, blocking contractile mechanisms in the lower urinary tract. This discovery of potential novel mechanisms of smooth muscle contraction provide the basis for bench to bedside #translational solutions for the #treatment and #care of #LUTS #BPH and #OAB by introducing novel #pharmaceutical options in the #future! Data are from our #research group focusing on basic and clinical research in #urological diseases, particularly benign prostatic hyperplasia, voiding dysfunction, prostate cancer, and minimally invasive surgery. Breaking Barriers Through Multi-Disciplinary Osteoarthritis (OA) Research - the OARSI - Osteoarthritis Research Society International World Congress in Vienna, April 18-21 2024 Scientists from Chondrometrics GmbH and Paracelsus Medizinische Privatuniversität (PMU) will deliver oral & poster presentations on advanced quantitative measurement of articular tissue pathology. The #OARSI congress (https://lnkd.in/gyqNm8aH) is the largest global platform of OA research. Attracting professionals from all disciplines in academia & industry, >1,000 attendees from >40 countries are expected in Vienna. OARSI is the premier society to advance research on #OA, a condition affecting >600 million people worldwide. Felix Eckstein, Professor at PMU & CEO at Chondrometrics recently became the Austrian Ambassador for OARSI. He enthusiastically anticipates the World Congress to be hosted in Austria, where his research group is affiliated with the Ludwig Boltzmann Institute Arthritis and Rehabilitation (#LBIAR), led by Tanja Stamm. On April 19 (8:45-10:15; Abstract No20) he is speaking on the analysis of MR images with and without blinding to acquisition order, and whether it impacts effect sizes of “#Sprifermin” on cartilage thickness in the FORWARD trial. A low-field magnet (0.55 Tesla) suitable for outpatient settings is compared with clinical (1.5, 3 & 7T) scanners, in collaboration with Frank W. Roemer, M.D.'s group & #Siemens Healthcare. Anna Wisser will present a poster (No207) on the impact of diabetes mellitus on physical function & thigh muscle strength in Osteoarthritis Initiative (#OAI) participants. The work is part of a broader project on metabolism and tissue function within the #LBIAR. Clemens Heistracher & Jana Eder contribute poster No084 on fully #unsupervised automated segmentation of thigh muscle & adipose tissue, without using labeled data, entailing important advances in automated segmentation technology. Wolfgang Wirth presents a poster (No491) on automated image segmentation, comparing different MRI sequences in detecting OA cartilage thickness change (co-authors Akshay Chaudhari & David Hunter). A 2nd poster (No493) on #sex-differences in radiographic knee joint space width & cartilage morphometry in size-matched subjects is part of a broader initiative, characterizing “sexual dimorphism” in tissue as risk factors for knee OA, recently submitted to a Special Issue of Osteoarthritis and Cartilage Journal.
