You're planning a statistical study. How do you ensure your sample size is large enough?

Without an appropriate sample size, the possibilities to develop an accurate analysis of the collected data are few. Let's assume that in an exam of a certain specialty we have only 5 to 10 students, is this enough to reach out at conclusions and predict the rate of success of a student? Of course not. And, furthermore, this reduced sample size, limits the use of many statistical or machine learning techniques used for modeling. Of course, we can use simulations, but even here the quantity and quality of the variables available from the sample size affects the simulation. We all agree "The more the better". Even sometimes "cleaning" and "selecting" the best observations is a very hard work!

More data is not always better. There is a trade-off between costs and benefits. Collecting more data is expensive, often increasingly so, because harder-to-reach respondents are, well, harder to reach (at increasing monetary or time cost). On the other hand, the benefit of more observations in terms of precision (lower variance) less than doubles when you double the sample size. Large-sample approximations are known to work well for sample sizes as small as 20 (for simple statistics such as averages). If you have funding for 1,000 observations, consider whether the precision of 500 observations is sufficient, and use the other 500 in another project. That way you can use your resources more efficiently.

The number of experimental units in an experiment/ clinical trial should always be large enough to provide a reliable answer to the question(s) addressed. It is an essential and crucial part of the design of any study. Sample size must be large enough to enable the detection of meaningful (engineering, clinical, biological, ...) effects.

Consider the following factors: 1-Determine the desired level of statistical power and significance level. 2-Define the effect size, which represents the magnitude of the expected difference or relationship. 3-Conduct a power analysis using appropriate statistical methods or software. 4-Consider the study design, such as the type of analysis and sampling method. 5-Ensure representative sampling to obtain a diverse and unbiased sample. 6-Consider the variability of the population to estimate the required sample size accurately. 7-Perform a literature review to identify similar studies and their sample sizes. 8- Balance practical constraints such as time, budget, and feasibility. 9- Consider stratification or sub-group analyses if applicable.

A small sample might unintentionally focus on specific groups, such as affluent urban households, resulting in biased conclusions. However, a larger and more diverse sample, encompassing various income brackets and geographic regions, offers a more reliable portrayal of the country's overall income distribution.

Several pieces of information are needed to estimate sample size, namely (not an exhaustive list): - type of trial - structure of the trial: number of arms (treatments), blocking variables, use of covariates, etc. - the primary endpoint on which the calculation is based together with the statistical test procedure. - objective : testing for equivalence, inferiority, superiority, etc. - direction of the statistical test: two-sided vs. one-sided - risk levels: acceptable confidence level and desired power - effect size - estimate of variability - drop out rate - etc.

Sample size in statistical studies is influenced by several factors with precision being one of them, When aiming for higher precision, a larger sample size is often required. For example, consider a humanitarian organization aiming to estimate the number of individuals affected by a natural disaster in a specific region. If the organization conducts a survey with a small sample size, the estimated number of affected individuals might lack precision. This imprecision could lead to inadequate resource allocation, resulting in either insufficient aid reaching those in need or unnecessary oversupply in certain areas.

Required sample size, and the power of any study plan, combines the study design with the expected effect. Power depends on the expected effect, sample size, design, and analysis choices that are valid for that design. You may wish to know the smallest parameter value that could be detected given power and a sample size, and this is also dependent on design and analysis. I refer only to choices which are appropriate for the design. If you randomized clusters, your analysis must take this into account, but if you assign units within clusters, you could choose fixed or random effects. Choices impact power and generalizability.

Several factors influence the determination of sample size 1. Effect Size: A larger effect size often requires a smaller sample size to detect the effect accurately. 2. *Significance Level (Alpha):A lower significance level (e.g., 0.01 instead of 0.05) requires a larger sample size. 3. Power of the Test:Higher desired statistical power (probability of detecting an effect if it exists) requires a larger sample size. 4. Variability or Standard Deviation: More variability in the data requires a larger sample size to achieve precision.

Simpler designs which have a clear scalar formula for the variance of the parameter make for simpler power and sample size estimates. More complex analysis without simple formulas often requires simulation procedures. Studies which are not using frequentist inference and instead are making Bayesian inferences are testing different hypotheses and thus have different methods to determine required sample sizes. Be sure to clearly understand your study goals before estimating the require sample for them. Many methods require assumptions about population parameters. Don't guess. Use the literature and available data to make educated assumptions, and clearly report them and why you think they are accurate.

One way to estimate the amount of data needed to obtain significant evidence of an effect of unknown size (induced e.g. by some intervention on the parameters of an industrial process) is to (i) create a model of the pre-intervention process, (ii) alter (to a variable extent) the parameters of this model to mimic the desired effect, (iii) run a certain number of simulations of the process under the altered parameter set and (iv) compare the distribution of the synthetic process output to the pre-intervention one, establishing whether any discrepancy between the two is statistically significant. In this way one can determine the required amount of data as a function of the size of the desired effect.

There are closed-form equations for determining sample size for simple experimental. As soon as the experimental structure becomes more complex, the use of dedicated statistical software such as nQuery may be needed. In complex cases, I typically run simulations in R.

One can also seek advice from experienced statisticians or experts in the field to ensure the chosen sample size aligns with the study's objectives and statistical requirements.

Nothing makes the expected impact increase like a budget meeting that concludes you cannot collect as much data as you may require. Rather than increased optimism on your interventions impact, put your energy towards alternative designs to detect your initial, smaller, likely more reasonable impact size. Be creative in your study design, not your expectations.

Selecting an appropriate sample size is crucial in statistical modeling to ensure the reliability of results. Consider the population variability, desired level of confidence, and acceptable margin of error. A larger sample enhances precision but may be resource-intensive. Employ power analysis to strike a balance. Define the effect size and significance level to estimate sample size accurately. Tailor the size to the study's specific goals and constraints. Ultimately, a well-considered sample size is the linchpin for robust statistical modeling, fostering confidence in research outcomes.

If looking at relationships and/or differences between/within variables then your sample size is directly related to your smallest effect size of interest (SESOI), your alpha and power. The SESOI is the smallest difference/relationship you care about in your research question. Large effects require smaller samples; small effects require larger samples. Given that the alpha and power are often determined by the field as ‘standards’, then deciding what effect size you would care about is really the key question in figuring out your sample size. In addition, consideration should be given to the feasibility of obtaining the required sample and so collaboration across research centres and labs is often advisable when large samples are needed.

Work to understand the community and populations that will participate in your study. Making assumptions about variance or outcome rates from rural Midwest areas for studies in Urban Mexico sounds silly, and it is. Just like effect size "shirt sizes" are stupid, so is any rule of thumb about population parameters. Humanity is so interesting and diverse, don't make bad science because of ignorance. If you truly have no idea about the basics of a population, perhaps you need to do an initial study to understand the basics before a randomized trial.

The sample size to use depends on the level of knowledge of a phenomenon. Fractional designs requiring fewer runs may be used in exploratory phases. Optimisation designs are helpful to fine tune the effects of influential factors and require an reasonable number of runs. Determining sample size must also take into account the data analysis plan. In complex statistical models such as generalised linear models optimal designs are used. The idea is to identify the most essential designs points to use to estimate the model parameters of interest with the smallest sample size possible. D-optimal (and variants) and used when the cost of experimentation is very high and or when testing is time consuming (aeronautics).
