Print your latest copy of the SOP, and hand it to your newest person to perform the work as it's written. Have them shadowed by your most senior operator. When the senior operator says "stop, that's not how we do it" you have changes to make. Then, your most experienced regulatory person looks at it and when they find a CGMP gap engage your change control system. Rinse and repeat. GMP rules to live by: 1. Say what you'll do. 2. Do what you said. 3. Document what you did. (If it's worth doing, it's worth documenting.) 4. If there's a difference between 3 and 1, start over at 1. 5. Get better. 6. Go back to 1.

In my experience, it is to align the GMP requirements to a standard and then to align the standard to an SOP. Once that action is taken, have another party confirm that the SOP is in alignment with the standard which in turn should be in alignment with the GMP requirements.

Implementation VS Culture Periodically observe that SOP is following as it is written or is there any deviation.It is a common observation that apart from having a SOP its implementation is not as per demand of SOP because people are following culture of the pharma.If we take example of working in cleanrooms, it is clearly mentioned to ensure door interlocking with alarm system but as per culture usually it is bypassed.So just having a written SOP is not the assurance of its implementation rather have to ensure it's implementation in true sense.

I would suggest that go through the requirements of regulatory authority and check the relevance of practices in your process design. Evaluate that how it is feasible to practice the regulatory needs in your system and where process need to be aligned according to that. Define the exact practices in the SOP and train employees to practices that regularly. Keep eyes on the regulatory updates and update your procedure accordingly. Train on the updates to the users and keep on practicing those things.
