In this application note, we show an example of 5-color panel on Platelet Poor Plasma (PPP) extracellular vesicles (EVs) isolated from healthy donors using the CytoFLEX nano flow cytometer. 👉 PPP EVs were prepared using centrifugation, filtration and size exclusion chromatography (SEC) using qEV single 70 nm (Izon). 👉 One fraction was selected based on the abundance of extracellular vesicles, ensuring sufficient dilution after staining, and removing antibody aggregates and other contaminants. 👉 The antibodies used were treated to eliminate aggregates by spinning at high speed, followed by titration to adjust for fluorescent spillover. 👉 Compensation, using the compensation matrix, was applied to the sample to account for any spectral overlap.
