Suleiman et al. presented two cases of locoregional recurrence of PIK3CA-mutated head & neck squamous cell carcinoma (SCC). After failure of chemotherapy, combinatorial immunotherapy led to better response. Kolck et al. showed that body composition analysis of preoperative computed tomography images of patients with intrahepatic cholangiocarcinoma is a better tool for risk stratification than body mass index. Wang et al. demonstrated DOT1L as a target of dexamethasone in B-lymphoma. Silencing expression of DOT1L reduced growth of B-lymphoma cells. Specifically, they showed that dexamethasone decreased DOT1L expression in dexamethasone-sensitive B-lymphoma cells & MLL-rearranged leukemia cells, but not in dexamethasone-insensitive acute monocytic leukemia cells. This suggested that DOT1L may be an underlying novel indicator of dexamethasone sensitivity against hematological malignancies. Yang et al. analyzed performance of a blood test for detecting hepatocellular carcinoma— dual-target HCC blood test (DT-HBT)—using GNB4 & Riplet methylation. Study suggested DT-HBT to be a reliable & high-performing blood test for diagnosis & monitoring of HCC. Adaranijo et al. applied grounded theory research method to examine the self-identities of women with breast cancer in Nigeria. The study suggests emergence of three self-identities: (1) valued self-identity before breast cancer; (2) dependent and determined self-identities during treatment; & (3) devalued self-identity post-treatment. Study may allow caregivers to understand profound perpetual psycho-emotional impact that breast cancer has on sufferers & survivors. Prior studies have shown that myeloproliferative neoplasm (MPN) with JAK2-V617F mutation has an increased expression of programmed death-ligand 1 (PD-L1) in megakaryocytes leading to cancer immune evasion by inhibiting T-lymphocytes. Recently, Ibrahim et al. compared PD-L1 expression in bone marrow (BM) of patients with JAK2-mutant & JAK2-negative MPN, with normal controls. Results suggest that patients with JAK2 mutation have a higher PD-L1 expression in megakaryocytes than that in normal controls. A combination of checkpoint and JAK2 inhibitors may be an active treatment option. Morita et al. conducted retrospective observational study on patients with EGFR-positive NSCLC with postoperative recurrence. Metastasis to the CNS & bone were poor prognostic factors. Study may potentially help clinicians to determine the timing of administering osimertinib to the patients. A review by Guo et al. discusses therapeutic potential of terpenoids in targeting mitochondria to induce cancer cell death. Terpenoids lead to apoptosis, pyroptosis, necroptosis, ferroptosis, autophagy, & necrosis caused by mitochondrial permeability transition. Wilson et al. present a case with metastatic non-myofibroblastic sarcoma with EML4-ALK fusion. First description tracking development of resistance mutations. The case study questions utility of presence of G1202R mutation as marker of #lorlatinib sensitivity in non-lung ALK rearranged tumors, contrary to experience in lung cancer.
