🚨 Indee Labs posted a manuscript that applies TIMING™ from CellChorus to show that CAR-T cells manufactured with the Indee platform have superior motility and resistance to activation-induced cell death (AICD). 📖 Background: Indee is developing Hydropore, a non-viral technology that utilizes microfluidic vortex shedding to deliver small molecules, nucleic acids, proteins, and gene-editing complexes to immune cells with low impact to cell viability and function. The technology is designed to bypass the challenges and limitations of viral transduction and traditional transfection via electroporation, allowing scientists to accelerate their research goals from discovery to manufacturing. 📋 The approach: The team compared hydroporation with nucleofection in the generation of CAR-T cells. 📈 Key findings from the research include: - Scaled-up hydroporation can process 5 x 10^8 cells in less than 10 seconds. - Hydroporation yields a higher number of transfected cells compared to both nucleofection and electroporation, partly because CAR-T made with hydroporation (Hy-CAR-T) robustly grew and divided. - Hy-CAR-T cells CAR-T from nucleofection (Nuc-CAR-T) in terms of T cell subsets, activation, and pro-inflammatory cytokine secretion. - Hy-CAR-T secreted higher levels of the anti-inflammatory cytokine IL-10, which is linked to better CAR-T function. - TIMING data showed that individual Hy-CAR-T target seeking. contact dynamics, and target killing were just as robust as Nuc-CAR-T. - TIMING measurements of cell movements during target engagement (synapse) showed that Hy-CAR-Ts maintained significantly more motility than donor-matched Nuc-CAR-T cells. 🚨CellChorus PUBLICATION ALERT - Nature Cancer publication identifies a clinically effective subset of CAR T cells by profiling with TIMING™ 📊 THE RESULTS: New research published in Nature Cancer by co-authors from the University of Houston, Kite Pharma (Gilead Sciences), the MD Anderson Cancer Center, Texas Children's Hospital / Baylor College of Medicine, and CellChorus applied TIMING analysis to identify a subpopulation of T cells (CD8-fit cells) that are migratory and capable of serial killing. The percent of CD8-fit cells within infusion products was correlated with complete clinical responses. ⚡THE SIGNIFICANCE: Chimeric antigen receptor (CAR) T cells can be a life-saving treatment for patients with cancer and other disorders. Despite this promise, not all patients respond to therapy and some experience severe side effects. It is therefore essential to identify recipients who are likely to benefit. 📄 THE APPROACH: Scientists conducted multifaceted single-cell analysis of CAR T cells from axi-cel infusion products of patients with diffuse large B-cell lymphoma, including dynamic single-cell analysis with the TIMING platform. 🔬 THE PLATFORM: TIMING applies AI+ML to quantify how individual immune cells move, interact, kill, survive, and secrete biomolecules in their interactions with target cells at single-cell resolution. Researchers from theSingle Cell Lab at the University of Houston have applied TIMING™ data to study redundancies in cytotoxic mechanisms employed by CAR T cells. Research published in Cell Death & Disease shows that leukemic targets are sensitive to multiple granzymes whereas solid tumors are sensitive to granzyme and Fas ligand.
