Protein kinases are among the most popular targets for drug development. To date, over 120 FDA-approved drugs are protein kinase inhibitors, with 42 of these approved in recent years and 7 of them in 2023. However, protein kinases pose significant challenges, particularly with regard to selectivity. Most FDA-approved drugs are ATP-competitive, or type I inhibitors, which can struggle with selectivity. One strategy to address this issue is developing compounds that utilize alternative inhibition mechanisms.
