    In anticipation of our upcoming attendance at the European Society of Gene and Cell Therapy (ESGCT) conference in Rome next month, I'd like to share some insights on gene editing strategies, with a particular focus on two trending transposases. The PiggyBac (PB) and Sleeping Beauty (SB) transposon systems are valuable tools for non-viral gene editing. Both systems recognize specific inverted terminal repeats (ITRs) that flank the gene of interest (GOI) on the transposon DNA. Although they share a similar function, PB and SB systems differ in several key aspects ⤵️ -𝐈𝐧𝐬𝐞𝐫𝐭𝐢𝐨𝐧 𝐒𝐞𝐪𝐮𝐞𝐧𝐜𝐞 PiggyBac: T̲a̲r̲g̲e̲t̲s̲ ̲T̲T̲A̲A̲ ̲t̲e̲t̲r̲a̲n̲u̲c̲l̲e̲o̲t̲i̲d̲e̲ sequences, and integration tends to be relatively random but is dependent on the presence of these specific sequences. Sleeping Beauty: T̲a̲r̲g̲e̲t̲s̲ ̲T̲A̲ ̲d̲i̲n̲u̲c̲l̲e̲o̲t̲i̲d̲e̲ sequences, with a preference for regions of active chromatin, meaning it often integrates into regions that are actively transcribed. -𝐑𝐞𝐯𝐞𝐫𝐬𝐢𝐛𝐢𝐥𝐢𝐭𝐲 PiggyBac: The excision of PiggyBac is precise and leaves ̲n̲o̲ ̲f̲o̲o̲t̲p̲r̲i̲n̲t̲, which is advantageous when reversible modifications are required. Sleeping Beauty: While the SB system can also be excised, it often leaves behind small DNA sequences (footprints) that may alter the original insertion site. -𝐂𝐚𝐫𝐠𝐨 𝐜𝐚𝐩𝐚𝐜𝐢𝐭𝐲 PiggyBac is known for its high transposition efficiency and ability to carry large DNA payloads, up to 100 kilobases. Sleeping Beauty is highly efficient as well, but it generally has a lower cargo capacity than PiggyBac, typically up to 10 kilobases. Very excited to be back to my hometown ROME for #ESGCT2024!
