The Health and Environmental Sciences Institute (HESI) is a nonprofit institution whose mission is to engage scientists from academia, government and industry to identify and resolve global health and environmental issues. HESI Vision: Creating science-based solutions for a sustainable, healthier world. HESI Philosophy: HESI 's scientific programs bring together scientists from around the world from academia, industry, and regulatory agencies and other governmental institutions, to address and reach consensus on scientific questions that have the potential to be resolved through creative application of intellectual and financial resources. This “tripartite” approach forms the core of every HESI scientific endeavor. As a non-profit organization, HESI provides a unique, objective forum for initiating dialogue among scientists with different perspectives and expertise. Industry sponsors provide primary financial support for HESI programs, but HESI also receives financial and in-kind support from a variety of U.S. and international government agencies. Quick History: HESI was established in 1989 under the scientific leadership of its inaugural Chair, Dr. Louis Lasagna, often referred to as the “Father of Clinical Pharmacology”. HESI initiated as an environmental and biomedical safety focused global branch of the International Life Sciences Institute. HESI’s first workshop on rodent liver tumors, in 1989, engaged fourteen scientific participants and was supported by a single staffer. As the organization grew in participation and scope, it opted to establish its own governance and programming. In 2002, HESI was recognized by the US Government as an independently-chartered and publicly-supported 501(c)3 tax exempt organization and has continued to grow and flourish in scientific scope and rigor. Today, HESI’s team of 10 scientific staff provide leadership to more than fifty scientific projects and programs that benefit human and environmental health.

🌿 New Advances in Botanical Safety 🌿 The HESI Botanical Safety Consortium’s Hepatotoxicity Working Group has published a proposed screening strategy to assess the hepatotoxicity and drug interaction risks of botanical supplements. With botanical products growing in popularity, ensuring their safety is more important than ever. 🔑 Key Highlights: 1. New Approach Methodologies (NAMs): Exploration of NAMs that are being proposed (in vitro and in silico models) to screen hepatotoxicity and botanical-drug interactions effectively. 2. Broad Botanical Testing: Testing a variety of botanicals through case studies, with assays evaluating cellular and molecular markers related to liver health. 3. Development of a Safety Toolkit: Creation of a toolkit to assist researchers in screening botanicals, informing further studies, and potentially reducing animal testing. Celebrating an inspiring Health and Environmental Sciences Institute (HESI)' #ProteinSafetyWorkshop, held last week at the Universidade do Porto in collaboration with Joana Costa and Isabel Mafra! 🎉 It was an honor to bring together over 20 brilliant speakers and moderators, whose expertise and insights helped us achieve the workshop’s objectives. The event brought regulatory perspectives from five distinct geographical regions worldwide. Discussions reflected on over 30 years of learnings from novel foods/feed safety assessments, how to address challenging cases like 'Intractable Proteins' and considerations for applying New Approach Methodologies (NAMs) in this space. One major recurring theme that emerged was the crucial need for clearer #criteria to support decision-making at various stages—a foundational element that will empower regulators and product developers alike to navigate novel product safety assessments with confidence. My heartfelt thanks to everyone who participated and helped make this event such a success. Looking forward to the continued #collaborations and steps forward we’ll take together!

Do you wonder how reproducible #onestep #RTddPCR is? Reverse transcription can introduce significant #variation in #geneexpression analysis (see for example this 2015 paper from Bustin et al. https://lnkd.in/eJKtexkG). So what about one-step RT-ddPCR? In this recent publication https://lnkd.in/evEd6h5M by the Health and Environmental Sciences Institute (HESI) CT-TRACS committee, Yasuda et al. aimed to detect undifferentiated pluripotent stem cells (#PSCs) in cell therapy products (#CTPs) to ensure biosafety. Their publication shows that on-step RT-ddPCR is: 🧬#reproducible: One-step RT-ddPCR was extremely reproducible across five sites, multiple markers, and multiple ddPCR instruments — all without normalization for input! 🧪#precise: Variation was mainly due to the #pre-PCR workflow. (In this case, it meant culturing #iPSCs, spiking them into cardiomyocytes, and performing RNA extraction.) ⚗#sensitive: One-step RT-ddPCR was exceptionally sensitive, detecting as low as 0.0003% (3 cells per million) residual iPSCs in CTPs. In conclusion, one-step RT-ddPCR demonstrates excellent reproducibility! #ddPCR #stemcells #digitalPCR #iPSC #ATMP #geneexpression #dPCR
