This week, we sequenced DLL1, short for Delta-Like Protein 1. DLL1 is an essential ligand for the Notch signaling pathway and is pivotal in embryonic development, maintaining adult stem cell populations, and ensuring proper tissue homeostasis. Therapeutically, DLL1 modulation offers promising avenues in oncology, as aberrant Notch signaling is implicated in many cancers, including T-cell acute lymphoblastic leukemia and breast cancer. Additionally, DLL1's influence on stem cell differentiation makes it a potential target in regenerative medicine, aiming to repair or replace damaged tissues. Our sequencing result demonstrates how Next-Generation Protein Sequencing™ data on the Platinum® instrument can be used to sequence and characterize potential therapeutic targets, such as DLL1. By analyzing the kinetic signatures from specific N-terminal amino acid recognizers, we identified 13 amino acids (F, Y, W, L, I, V, R, A, S, N, Q, E, and K), enabling identification of 4 peptides of DLL1 as shown in the figure. This week, we sequenced B2MG, short for Beta-2-Microglobulin. B2MG is a component of the major histocompatibility complex (MHC) class I molecules, which are involved in the presentation of peptide antigens to the immune system. It plays a critical role in immune surveillance by helping to display intracellular proteins on the cell surface for recognition by cytotoxic T cells. Elevated levels of B2MG are associated with chronic kidney disease and certain malignancies, making it a potential biomarker for these conditions and a target for therapeutic intervention. Our sequencing result demonstrates how Next-Generation Protein Sequencing™ data on the Platinum® instrument can be used to sequence and characterize potential therapeutic targets, such as B2MG.
