When we started Ramona Optics Inc. several years ago, we were fortunate enough to be awarded a Small Business Innovation Research (SBIR) grant from the National Institutes of Health (NIH ORIP). Whole-Brain Imaging of Neuronal Activity in Zebrafish … Scientists at the Howard Hughes Medical Institute studied live zebrafish larvae using a novel approach that combined genetics, calcium imaging, and light-sheet microscopy to observe whole brain activity. This method allowed scientists to record the activity of more than 80% of the 100,000 neurons in the zebrafish brain at single-cell resolution. Here is how it works (from journal Nature): The imaging system involves using genetically engineered zebrafish. The fish's neurons make a protein that fluoresces in response to fluctuations in the concentration of calcium ions, which occur when nerve cells fire. A microscope sends sheets of light rather than a conventional beam through the fish's brain, and a detector captures the signals like a viewer watching a cinema screen. I so am pleased to announce our new paper (https://lnkd.in/eRY_yUXQ), led by Sebastian Gutsfeld, showing that PFOS exposure causes visual startle response hyperactivity in zebrafish via a ppard-dependent mechanism. This was a labor of love, and a herculean effort, started at US EPA and painstakingly completed at the UFZ. This is the first time we chased down a causal mechanism, linked to a behavior phenotype, and was a follow-up to our earlier work, showing that structurally similar PFAS cause dark phase hyperactivity (https://lnkd.in/eYBzNK7).
