SiriusXT is delighted to collaborate with Tescan, a leading global producer and supplier of electron microscopes, to demonstrate the benefits of integrating soft X-ray microscopy into correlative imaging workflows that employ light and electron microscopy.
Tescan’s Martina Zánová will present initial findings from this collaborative project at EMC2024, which will take place from August 25th to 30th in Copenhagen. EMC2024, Europe’s largest event dedicated to microscopy and imaging, will take place at Bella Center, Center Boulevard 5, 2300 Copenhagen.
Soft X-ray Tomography (SXT) is a stand-alone, high-throughput, imaging technique that images the whole structure of intact biological cells with high natural contrast, without needing to stain or slice the cell. It can also be used as a pre-screening tool in correlative workflows by using it to target region of interest (RoI) that can subsequently be imaged by electron microscopy.
In the correlative imaging workflow conducted with Tescan, the RoI in the cell, identified by soft X- ray microscopy, was isolated into an in-situ lamella by removing unwanted cell material from above and below the RoI using Focused Ion Beam (FIB) milling. The exposed lamella was then moved to a cryo-transmission electron microscope (cryo-TEM) to image the RoI in the lamella.
