🔬 Optimizing Primary Antibody Dilution in IHC 🧫 In Immunohistochemistry (IHC), precision matters. Achieving the right balance between signal and background staining is key. The primary antibody, whether monoclonal or polyclonal, is a critical player in this process. Monoclonal antibodies, specific to a single epitope, excel in precision. Polyclonal antibodies, with the ability to bind various epitopes, offer sensitivity. So, how do we find the sweet spot? Titration: Start with the recommended dilution but don't stop there. Test more concentrated and less concentrated dilutions flanking the recommendation. It's a balancing act to achieve optimal staining. Positive Control: Use appropriate positive controls to fine-tune your dilutions. The right control tissue is your guide. Secondary Antibody: Experiment with different dilutions, especially when coupled with the Antigen Retrieval (AR) method and chromogen. For a starting point, 1 to 5 μg/mL is often recommended for the initial titration.
