Antibody Therapeutics (indexed by ESCI and Scopus, 2023 CiteScore: 8.7) just published a new article (currently in accepted manuscript format). The authors from Biologics Innovation Institute, Shanghai Jemincare Pharmaceuticals Co., Ltd. discovered a common light chain bispecific antibody targeting PD-1 and PD-L1 with favorable biophysical properties and potent efficacy. Notably, they have developed a unique and flexible H2PtY (Hybridoma-to-Phage-to-Yeast) platform for the generation of common light chain bispecific antibodies targeting a given pair of targets. This excellent, brand new (posted just hours ago) paper by Karunaratne et. al. describes studies on understanding the effects of site-specific light chain conjugation on antibody structure using hydrogen exchange-mass spectrometry (HX-MS). Quoting from the abstract: "Antibody drug conjugates (ADCs) represent one of the fastest growing classes of cancer therapeutics. Drug incorporation through site-specific conjugation in ADCs leads to uniform drug load and distribution. These site-specific modifications may have an impact on ADC quality attributes including protein higher order structure (HOS), which might impact safety and efficacy. In this study, we conducted a side-by-side comparison between the conjugated and unconjugated mAb. In the ADC, the linker-pyrrolobenzodiazepine was site specifically conjugated to an engineered unpaired C215 residue within the Fab domain of the light chain. Differential scanning calorimetry (DSC) and differential scanning fluorimetry (DSF) indicated a decrease in thermal stability for the CH2 transition of the ADC. Size exclusion chromatography (SEC) analysis, showed that conjugation of the mAb resulted in earlier aggregation onset and increased aggregation propensity after 4 weeks at 40 °C. Differential hydrogen-exchange mass spectrometry (HX-MS) indicated that upon conjugation, light chain residues 150-155 and 197-204, close to the conjugation site, showed significantly faster HX kinetics, suggesting an increase in backbone flexibility within this region, while heavy chain residues 32-44 exhibited significantly slower kinetics, suggesting distal stabilization of the mAb backbone."

Presumably associated with this year’s Nobel Prize in Chemistry regarding AI & protein/antibody, although this paper was published just in this month in Antibody Therapeutics, it already quickly becomes one of the most read papers. The authors of this new open access article from Just - Evotec Biologics describe a novel, cost-effective and accelerated approach to therapeutic antibody discovery, that couples de novo human antibodies derived in silico with high throughput screening technologies. Just-Evotec Biologics’ platform’s applicability is demonstrated here by the identification of a panel of humanoid antibodies that are effective and with high developability potential. At BTI, biologics discovery and development is a team effort. The Antibody Development Platform (ADP), spearheaded by Associate Professor Andre Choo, spans multiple core capabilities within BTI to accelerate the discovery and development of monoclonal antibodies. ADP starts by discovering novel monoclonal antibodies that target unique conformational epitopes and post-translational modifications, such as aberrant glycosylation only on cancer cells. This is achieved using a whole-cell immunisation approach. Unlike raising antibodies against a target protein, this phenotypic approach is target-agnostic and thus enables us to generate antibodies against specific cell types of interest. For example, the parental antibody of EBC-129 was developed in collaboration with clinicians to discover new antibody therapeutics for the treatment of Gefitinib-resistant non-small cell lung cancer. The antibody was raised against drug-resistant cancer cells and was put through various screening, target specificity, prevalence, modality screening and engineering. Once the lead candidate was identified, the pre-clinical material was produced and assessed for developability. Subsequently, the antibody went for further pre-clinical evaluation and development. In business news, Triveni Bio announced a $115 million Series B to support pipeline expansion. The round was led by Goldman Sachs Alternatives with significant participation from additional new investors Fidelity Management & Research Company and Deep Track Capital, and Series A investors Atlas Venture, Cormorant Asset Management, OrbiMed, Viking Global Investors, and Invus. Triveni plans to submit an Investigational New Drug (IND) application for its lead candidate, TRIV-509, in the first quarter of 2025. TRIV-509 is a monoclonal antibody inhibitor targeting kallikreins 5 and 7 (KLK 5/7) that has demonstrated superior efficacy compared to IL-4R inhibition in multiple preclinical atopic dermatitis models. TRIV-573 combines KLK 5/7 inhibition with a well-established IL-13 mechanism. For more information, read on… Recently, we have seen the role of #HybridLCMSMS in the analysis of #AntibodyDrugConjugates (#ADC) increase due to the complexity and number of #bioanalytical assays needed to support this modality in the #DrugDevelopment process. Antibody drug conjugates tend to consist of a toxic small molecule payload attached via a linker to an #antibody. The application of these drugs had mainly been in #oncology where their mode of action is for the antibody to attach at the desired target site with subsequent release of the small molecule to combat #TumorCells. Having a therapeutic entity with multiple distinct components means that we need to have analytical methodologies for various components. Gene&Cell Therapy >> BioNTech pays $20M upfront for two monoclonal antibodies from China's WuXi Biologics: BioNTech is paying out $20 million upfront to another Chinese partner in exchange for the rights to develop two preclinical monoclonal antibodies, this time with CRDMO WuXi Biologics. The deal also includes research, development, regulatory and commercial milestone payments, plus tiered royalties for the Shanghai-based company, which will use its antibody discovery technology platforms to discover the antibodies. While the targets of the latest deal with WuXi are still undisclosed, BioNTech has been building up its presence in the cancer space after making a name as a top Covid mRNA vaccine developer, with TD Cowen’s Yaron Werber noting in November that BioNTech has assembled “one of the most extensive” oncology portfolios in biotech. During BioNTech’s Innovation Day in November, executives spent hours discussing data from a variety of antibody-drug conjugates, bispecifics, CAR-Ts and vaccines for cancer. The German biotech paid $55 million upfront to collaborate with Biotheus in November in return for the global ex-China license of PM8002, a bispecific simultaneously targeting PD-L1 and VEGF now being tested in Phase II studies in China. Biotheus could also get more than $1 billion more in milestone payments. BioNTech also partnered with Biotheus in July for a preclinical bispecific and a clinical-stage monoclonal antibody for undisclosed cancer targets in exchange for an upfront payment. Then in September, BioNTech started a Phase III trial for its antibody-drug conjugate aimed at patients with hormone receptor-positive, HER2-low breast cancer whose disease has progressed on at least two lines of prior endocrine therapies or within six months of first-line endocrine therapy and CDK4/6 inhibitor in the metastatic setting.
