【Piperidine/Azetidine Scaffolds】 Piperidine and azetidine are usually used as a scaffold of bioactive compounds. They were used as an important scaffold of ACT-1016-0707, potent and selective lysophosphatidic acid receptor subtype 1 receptor (LPAR1) antagonist [1, 2]. The diverse nature and the chemical modification of #oligonucleotide therapeutics leads to the requirement for multiple separation techniques to characterize the quality attributes. This new data discussed two common chromatographic approaches IP-RP and SAX being used as orthogonal methods to analyze short oligonucleotides ranging from 15-40 mer. The mixture of sample with 5 base difference each was successfully separated on the Proeteomix SAX NP5 4.6 x 250 mm column via ion exchange mode based on the length of oligos (left figure), while the same sample was also well separated by the Bio-C18 300 Å column via Ion pare mode with TEAA as ion pair regent (right Figure). On both phases, the sample were eluted according to the length of the oligos, reflecting the stronger interaction of longer oligo with both IEX and RP resin surface of the columns.
