We’re funding vital research on myotonic dystrophy! 💡 Dr. Ami Ketley at the University of Nottingham is using cutting-edge techniques and computer analysis to study different tissues in mice with the same genetic info as people with Myotonic Dystrophy Type 1. By examining the heart, brain, and muscles, this research aims to uncover what’s really happening in these tissues. Understanding this could be crucial for tackling DM1. Happy to share the latest pre-print from my PhD work with the James Ellis lab, in collaboration with the Julio Martinez-Trujilo lab: https://lnkd.in/guTjSy3j Here, we used in vitro multielectrode arrays and pharmacological manipulations to characterize how functional connectivity and network-level firing properties were altered in cultures of human iPSC-derived SHANK2 neurons. We found that the activity generated by SHANK2 networks was hyper-synchronous compared to isogenic control networks, indicating enhanced functional connectivity. This network hypersynchrony was fully rescued by the chronic treatment of cultures with the group 1 mGluR agonist DHPG. We also get into some interesting analysis of intra-network burst firing dynamics, reverberating superbursts, and machine learning classification analysis using different network activity features.

Spinal cord nerve cells learn and remember completely independent of the brain The researchers carried out studies to determine how the spinal cord recovers from injuries by exploring how the nerve connections are wired, and how they function and change when we learn new movements. Part of the problem is the difficulty in directly measuring the activity of individual neurons in the spinal cord in animals that are not sedated but awake and moving. The team took advantage of a model in which animals train specific movements within minutes. In doing so, the team uncovered a cell type-specific mechanism of spinal cord learning. To check how the spinal cord learns, the lab built an experimental setup to measure changes in movement in mice, inspired by methods used in insect studies. “We evaluated the contribution of six different neuronal populations and identified two groups of neurons, one dorsal and one ventral, that mediate motor learning.” "These two sets of neurons take turns," explains the author. "The dorsal neurons help the spinal cord learn a new movement, while the ventral neurons help it remember and perform the movement later." #ScienceMission #sciencenewshighlights https://lnkd.in/grEMKPRv
