To transfer or not to transfer? The dilemma of mosaic embryos is discussed in this new narrative review by Elkin Muñoz, Fernando Bronet, Belen Lledó, Gabriela Palacios Verdu, Lorena Martinez-Rocca, Signe Altmäe, and Josep Pla, representing the Special Interest Group in Reproductive Genetics of the Spanish Society of Fertility. The review summarizes current knowledge about mosaic embryos in the field of assisted reproduction techniques, and is intended to serve as reference material for practitioners in reproductive medicine who must manage a mosaic embryo result after preimplantation genetic testing for aneuploidies. Based on the assessment of current evidence in this paper, the authors suggest that embryos with a result within the mosaic range should not systematically be discarded or disregarded for transfer as this practice could negatively affect the cumulative live birth rate per cycle. Unraveling the Genetic Origami: Insights into DNA Folding Variations in Sperm Cells Across Vertebrates Much like the intricate art of origami, DNA undergoes complex folding within the nucleus of a cell. The compaction of DNA varies across different cell types, such as somatic and germ cells, and is meticulously regulated by cohesin, a protein complex responsible for maintaining sister chromatid cohesion and facilitating DNA looping. This study reveals that in frog and human sperm, genomic DNA doesn't fold through cohesin, unlike somatic cells. Instead, specialized sperm proteins called protamines densely package the DNA in these cells. Previous research in mouse sperm had suggested cohesin's involvement, but a related study in this issue disproves that, attributing the data to chromatin contaminations from somatic cells. Collectively, these findings propose that genomic DNA packaging in sperm across all vertebrate species may differ significantly from the folding observed in somatic cells. This study by Tianli Chang and team at Xiangya Hospital of Central South University, Hunan, found that a novel identified homozygous nonsense variant of AK7 may be associated with MMAF-related asthenoteratozoospermia. The functional associations between mitochondria and sperm flagellar assembly observed in this investigation provide evidence for potential mutual regulation between AK7 and flagella-associated proteins during spermatogenesis. These findings not only enhance understanding of the complex molecular mechanisms governing sperm development, and underscore the importance of mitochondrial involvement in this critical process, but also expand the range of pathogenic variants related to MMAF. Discover SCOPE: The Comprehensive Genetic Study of the Embryo What does SCOPE assess? (3/6) Did you know that SCOPE can also determine the meiotic/mitotic origin? Another feature of SCOPE is its ability to detect the stage at which the chromosomal segregation error occurred, whether the aneuploidies happened during meiosis (the formation of eggs and sperm) or during mitosis (the early cell divisions of the embryo). Knowing the origin of aneuploidy can help better understand the prognosis and associated risks with the embryo, enabling informed decisions about its possible transfer. Additionally, it provides crucial information for genetic counseling of the parents. Male infertility is a significant factor in conception challenges, with semen analysis playing a crucial role in evaluating it. Teratozoospermia, indicating sperm shape abnormalities, can greatly impact fertility. A trend of teratozoospermia (over 96% abnormal sperm shapes) is on the rise according to our in-house data and one such unique case includes coiled tails. A couple came to our clinic for fertility evaluation and upon semen analysis, coiled sperm abnormality was seen in the majority of the sperms. upon evaluation, we did the whole exome sequencing to identify the genetic cause in this case. Excitingly, WE ARE THE FIRST TO IDENTIFY A MUTATION IN THE SPEM1 GENE CAUSING COILED TAILS IN HUMAN SPERM. Explore our groundbreaking findings in the field of andrology and embryology.
