Determining the appropriate dosing regimen and predicting ADC behaviour in humans is crucial to their clinical success. Challenges include: -Understanding the interplay between the antibody, linker, and cytotoxic payload & accurately predicting how the ADC will distribute and release the drug in target tissues versus non-target tissues -The need for special considerations for dose- and exposure-response analyses given the selectivity of the antibody and the potency of the cytotoxic payload - Navigating the requirement for more rational dose optimization strategies off the back of Project Optimus guidelines which may be more time consuming & costly, particularly for smaller biotechs As the ADC Pharmacokinetics & Clinical Pharmacology Summit will unpack these challenges in under a month, I wanted to highlight a final opportunity to save up to $350 on registration, expiring this Friday (09/13) Register HERE: https://ter.li/5uu6ht Our industry-leading speaker faculty includes: Alison Betts, Doug Leipold, Faye (Zufei) Zhang, Frank Christian Dr. Richard, Hsuan Ping Chang, Ilse De Salve, Jeff Kearns, Jianzhong Wen, Li Li, Michael Z. Liao, PhD, MLS, Niyanta Kumar, Rajeev Menon, Sandhya Girish, Seema K., Venkatesh Pilla Reddy, Xiaogang Han, Yuting Wang, Ph.D, Zuzana Antosova Looking forward to meeting everyone in Boston! 😊

Organoid co-cultures and organ-on-a-chip (OoC) systems are advanced in vitro models that further enhance the physiological relevance of organoids by incorporating a broader array of tissue components compared to conventional organoid cultures. Read this mini review to learn about the Predictive Power of Organoids in Drug Discovery and see key takeaways below: - Although immortalized cell lines and animal models are useful for many applications in drug discovery, they are limited by their reductive and cross-species characteristics, respectively, which affect their ability to predict human in vivo responses with sufficient accuracy. - Organoids are high-fidelity biological models that can recapitulate the cellular structure and function of specific organs, can be expanded and stored for long periods, and are compatible with a wide range of assays, including high-throughput screening. - Organoids have demonstrated predictive power with notable sensitivity and specificity in recent drug evaluation studies. - Incorporating human stem cells or stem cell-derived organoids into complex systems such as co-culture and OoC systems is a powerful way to closely model metabolic, transport, and other physiological cellular characteristics of the tissue of interest, maximizing the predictive value of these systems for drug discovery research.
