🔬 Delighted to announce the successful completion of an enriching internship at the Centre for Plant Biotechnology and Molecular Biology, Kerala Agricultural University, Thrissur. From June 5th to July 4th, 2024, I had the privilege to delve into Molecular Biology Techniques for undertaking mini project in Biotechnology. ⚜ The internship focused on conducting Fidelity Analysis of plant samples- specifically Plantain Banana and Black Pepper. Key skills acquired include: ◼ Plant Genomic DNA Isolation: Mastered the CTAB method and preparations of CTAB buffers, CTAB solution, TE buffer and EDTA solution. ◼ Nanodrop Dilutions: Utilized for precise assessment of DNA purity and concentration in samples, ensuring the integrity of genetic material for accurate analysis. ◼ Agarose Gel Electrophoresis: Essential for molecular diagnostics, this method enabled me to detect, identify, and quantify nucleic acids within plant samples, facilitating detailed molecular analysis. ◼ Gel Documentation System: Through the use of a Gel Documentation System, I learned to effectively visualize, analyze, and document results obtained from gel electrophoresis. This capability enhanced my ability to interpret experimental outcomes with precision and clarity. ◼ PCR (Polymerase Chain Reaction): Applied in fidelity analysis, PCR involves precise temperature cycling for denaturation, annealing, and extension phases. The customization of annealing temperatures for various primers optimizes specificity in target amplification, critical for discerning genetic fidelity among Plantain Banana and Black Pepper samples. ◼ ELISA (Enzyme-Linked Immunosorbent Assay): Used for quantifying antibodies or antigens in samples. I also learned to prepare Coating buffer, substrate buffer, and PNPP solution for ELISA. This technique enhances understanding of plant immune responses. ◼ Media Preparations: Prepared PDA Media for fungal cultures, LB broth for bacterial cultures and King's B Medium for specialized bacterial growth. ◼ Bacterial Genome Isolation: Techniques in bacterial genome isolation were explored, emphasizing the extraction and purification of bacterial DNA for genomic studies. I'm deeply grateful to Kerala Agricultural University for this enriching opportunity and to my mentors for their continuous support and guidance.
