PolyVation completes qualification of 100 L UltraFiltration unit. We recently completed the qualification of a new 100 L UltraFiltration unit for the purification of GMP-compliant polymers for use in demanding medical products. UltraFiltration (also Tangential Flow Filtration - TFF) is a membrane separation technique that is used to purify polymers by removing unwanted impurities over a semi-permeable membrane, while retaining the polymer. UltraFiltration is a faster and more scalable technique than classical dialysis. The unit was designed based on elaborate User Requirement Specifications that were composed after extensive Development and Engineering work with our smaller-scale R&D (picture) and GMP UltraFiltration units. The scaled-up system provides enhanced control over temperature, pressure and volumes, resulting in reduced processing time and even higher purity of the polymer. After purification, the UltraFiltration unit also enables further concentration of the polymer solution in order to reduce volume prior the next processing step, often freeze-drying. The system is supported by in-line impurity monitoring and can run largely automated. After Factory Acceptance Testing at the equipment supplier, the unit was delivered to PolyVation, where it was qualified after comprehensive Site Acceptance Testing and IQ/OQ tests. The UltraFiltration unit will be put in use shortly, as soon as the construction of new clean production areas is finalized (see previous post). In parallel, also new plate freeze-dryers are being installed to remove the water from the purified solution and subsequently the dried polymers to be packaged under controlled conditions. This is another milestone that underlines PolyVation’s growth and commitment to support our customers’ increasing needs for the highest-quality GMP-grade polymers. The polymer dissolving process is called polymer solubilization, and a complete solubilization is essential to enhance dispersibility, emulsification ability, and particle suspension.
