(1/3) The main types of cell-penetrating peptides (CPPs) based on their chemical and structural properties are cationic, anionic, amphipathic, and hydrophobic peptides. Cationic CPPs are mainly composed of basic amino acids such as arginine and lysine, while amphipathic CPPs have a structured arrangement of polar and nonpolar amino acids. Hydrophobic CPPs are typically composed of nonpolar amino acids and interact with the hydrophobic region of the cellular membrane. Anionic CPPs, which are negatively charged, are also mentioned in the document, but they are not explicitly described as a distinct category based on their chemical and structural properties. Improved Clearance of Host Cell Protein Impurities at the Polishing Purification Step Using Multimodal Chromatography In this paper, we studied a novel therapeutic protein molecule which had a high level of HCP co-purification throughout the downstream process. Here, we focused on the polishing purification step and developed an effective strategy for improving HCP clearance using multimodal chromatography (MMC) resin, Nuvia cPrime. A high throughput process development (HTPD) workflow was used to identify the resin and process conditions which could enable significant HCP clearance while maintaining acceptable product quality and process performance. This strategy was highly effective and enabled 80% reduction in total HCP content, including some problematic and difficult to remove candidates such as Peroxiredoxin-1, Serine protease HTRA1, Clusterin and Lipoprotein lipase. https://lnkd.in/gSVJGcvw #aspenalert #biotech #bioprocess

If you are doing peptide mapping and want 100 percent coverage of tertiary or secondary protein structures, try using multiple digestion enzymes with different cleavage sites, make sure the pH is optimal for the enzymatic reaction without causing chemical changes of primary analyte structures, and higher temps over longer run times(50 degree celsius over shallow up to a half day long gradient) may lead to full coverage. Obviously you could switch up column size and fgroup, flow rates, gradient curve, mobile phases, acidic modifier etc., however these will all have significantly less impact than the pH and enzymes and temp/timing of the run. #separations #chemistry #method #development #scientist #variables #variability #coverage #peptidemapping #tertiarystructure #peptides #peptide #digestion #cocktailofenzymes
