Thrilled to share an exciting update from our NIH-funded research! Our interdisciplinary approach is paving the way for groundbreaking glaucoma therapy. Our team seamlessly combines stem cell differentiation, high-throughput drug screening, cutting-edge cell biology techniques, and animal models in our quest to discover a novel drug for glaucoma therapy. The promising news? Our newly identified drug is showcasing significant axon regeneration in the optic nerve of treated mice post optic nerve crush (ONC) injury. The growth of axons is vividly demonstrated through immunohistochemistry targeting the growth-associated protein 43 (GAP43). In glaucoma, where retinal ganglion cell axons degenerate in the optic nerve, connecting our eyes to the brain for vision, this discovery offers a potential pharmacological agent to promote optic nerve regeneration. Here's the kicker: The compound is already FDA approved for diagnostic purposes, marking a crucial step forward. Next on our agenda? Human studies! Molecular analysis can uncover chromosomal aberrations that can aid in detecting melanoma, but traditional methods for detecting copy number variation can be inefficient and costly. This study demonstrated the use of Droplet Digital PCR technology for detection of copy number variation in a commonly deleted tumor suppressor gene in melanoma. The data showed that ddPCR assays could identify deletions in melanocytic tumors with high sensitivity and specificity, suggesting potential for novel differentiation in histologically ambiguous cases. We're excited to share our collaborative effort on a new case report, published in partnership with the Department of Pathology and Molecular Medicine, Second Faculty of Medicine, Charles University, and Motol University Hospital, Prague. This report delves into the rare case of a CIC::NUTM1 sarcoma of the brain in a 6-year-old patient. This type of sarcoma is particularly aggressive and often affects the axial skeleton. Our findings contribute to the growing body of knowledge on this rare disease, expanding on previous reports and highlighting the importance of accurate diagnosis and timely intervention. We hope this research will lead to improved understanding and treatment of CIC::NUTM1 sarcomas, ultimately benefiting patients with this challenging disease. Adipose tissue organoids are still vastly underrepresented in the field of 3D in-vitro biology. Fat tissue is often neglected in the organ world. But as I learned from Dr. Elvira Weber, it is a full organ. And the most communicative organ at that. No matter the pathology you are looking at, you will find a connection to adipose tissue. From the more obvious, like cardiovascular disease, to cancer, Alzheimer’s, Parkinson’s, and even blindness. Fat surrounds everything. It's involved everywhere.     Dr. Weber and I sat down a few weeks ago to discuss her pioneering work with adipose tissue organoids, or “Adipoids” as she calls them. Her work is foundational to 3D in-vitro work with adipose tissue.
