Thank You Labiotech.eu for hosting this podcast. In this almost 30 min I had the opportunity to talk about the exciting clinical development of ISTH0036 - a TGF-beta 2 targeting antisense. This is the lead compound of Isarna Therapeutics GmbH, currently in Phase 2 trials for wet AMD and DME (with positive data just presented at @OIS and @ARVO by our CMO, Prof. Marion Munk, M.D., Ph.D ...). I also discuss briefly the promising antisense technology and its use as clinical therapy in general. Check it out!

Have you heard of PsCas9? 🤔 Identified bioinformatically, 𝘗𝘢𝘳𝘢𝘴𝘶𝘵𝘵𝘦𝘳𝘦𝘭𝘭𝘢 𝘴𝘦𝘤𝘶𝘯𝘥𝘢 Cas9, or PsCas9, is a type II-B Cas9 ortholog found in the human microbiome. With off-target editing being a high focus when developing therapeutics, the group developed a method dubbed '𝗗𝘂𝗽𝗹𝗲𝘅-𝗦𝗲𝗾' to effectively detect off-targets with high sensitivity and demonstrated PsCas9 has great fidelity and could be a safer option compared to SpCas9 for developing therapeutics. Characterising bacterial extracellular vesicles as intranasal postbiotics: in their latest work, Agnieszka Razim, Irma Schabussova and collaborators aimed to analyse EcO83-derived extracellular vesicles and explore their interactions with host cells, emphasising their potential as postbiotic therapeutics. EcO83-EVs were isolated, purified, and characterised in accordance with the MISEV guidelines. https://lnkd.in/eyfuzR-6 Ex vivo studies using human nasal epithelial cells revealed that EcO83-EVs enhanced the expression of proteins associated with oxidative stress and inflammation, suggesting a strong and effective interaction between the EVs and host cells. 🌟 Excited to share the Editor’s Choice Paper: Deciphering Metabolic Pathways in High-Seeding-Density Fed-Batch Processes for Monoclonal Antibody Production: A Computational Modeling Perspective. 🥼 This paper is authored by Carolin Bokelmann, Alireza Ehsani, Jochen Schaub and Fabian Stiefel. 💡 Due to their high specificity, monoclonal antibodies (mAbs) have garnered significant attention in recent decades, with advancements in production processes, such as high-seeding-density (HSD) strategies, contributing to improved titers. This study provides a thorough investigation of high seeding processes for mAb production in Chinese hamster ovary (CHO) cells, focused on identifying significant metabolites and their interactions. We observed high glycolytic fluxes, the depletion of asparagine, and a shift from lactate production to consumption.
