Detecting single nanoparticles and viruses by combining microfluidic laminar flow and confocal fluorescence microscopy: in their latest work, Paz Dor, Eitan Lerner at The Hebrew University of Jerusalem and collaborators introduced a rapid method for detecting single nanoparticles using a confocal-based flow virometer, combining microfluidic laminar flow with correlated fluorescence signals from free dyes and labelled antibodies to determine nanoparticle volume and specificity https://lnkd.in/e2ap-cSm The authors further demonstrated that hydrodynamic focusing significantly improves assay sensitivity, enabling the detection of viruses at clinically relevant concentrations. Characterization of the bone marrow adipocyte niche with three-dimensional electron microscopy - Amira Software Use Case Unlike white and brown adipose tissues, the bone marrow adipocyte (BMA) exists in a microenvironment containing unique populations of hematopoietic and skeletal cells. To study this microenvironment at the subcellular level, we performed a three-dimensional analysis of the ultrastructure of the BMA niche with focused ion beam scanning electron microscopy (FIB-SEM). This revealed that BMAs display hallmarks of metabolically active cells including polarized lipid deposits, a dense mitochondrial network, and areas of endoplasmic reticulum. Interesting LNP data coming out of the Thomas L. Andresen lab at the Technical University of Denmark. 💥 Roughly only 12% of mRNA-LNPs are "empty" ⭕ This value is even lower for siRNA (5.8% empty) and much higher for plasmid DNA (26.4% empty), indicating the cargo size plays a major role in the amount of cargo encapsulation. They tested this across four well known LNP standards and obtained similar results. 𝐂𝐨𝐧𝐬𝐞𝐪𝐮𝐞𝐧𝐜𝐞𝐬? A vast majority of LNPs in clinically relevant formulations are encapsulating nanomedicines and are thus able to exert their therapeutic activities.
