Cyclosporine A (CsA), a potent immunosuppressive drug used to treat autoimmune diseases and prevent organ transplant rejection, requires high purity for safety and efficacy. Impurities can lead to side effects and toxicity. Recent advancements in High-performance liquid chromatography (HPLC) have improved CsA purification, but challenges remain in achieving purity levels above 95%. This study employed a multi-step HPLC method to enhance CsA purity. After exploring various HPLC conditions, including mobile phase solutions and temperature, the optimal method was determined. Using acetonitrile-water-tert-butyl methyl ether-phosphoric acid (430:520:50:1) as the mobile phase, with a column temperature of 70 °C and specific chromatography column specifications, an average CsA yield of 80.1% with 97.2% purity was achieved. Surface-enhanced transmission Raman spectroscopy (SETRS) can be employed to detect impurities in complex matrices, study shows. Paving the way for new quantitative applications. Currently, the US Pharmacopeia recommends liquid chromatography for the quantitative determination of 4-aminophenol (4-AP), a degradation impurity of paracetamol. However, the study’s authors targeted to explore SETRS, a spectroscopic technique that is more time-efficient and eliminates the need for organic solvents typically required in separation methods. Preparation Techniques of Polypeptide - Physical Extraction method. It is a new concept proposed in recent years. This method does not need any chemical #reagents when preparing the #peptides. It is based on an ultrahigh-pressure continuous cell flow instrument, which can break up more than 90% of E. coli cells. Therefore, after breaking by the instrument, all animal or plant tissue has been lysed, the cell wall and cell membranes have been ruptured, and active #ingredients in cells have been released and become peptides by continuous cell flow lysing. This method ensures the biological activity of the polypeptide to the greatest extent, but it requires high equipment with a single processing volume and low polypeptide yield, so it is not suitable for industrial production. #biopharma #biological #physical #biopharmaceutical https://lnkd.in/gRM9Xurm
