The generation and accumulation of α-Synuclein amyloid fibrils in the brain is a major pathological hallmark of Parkinsons´s disease. Accordingly, inhibition of fibril generation could be a viable treatment strategy. In this AppNote potential inhibitors of α-Synuclein fibrilization were evaluated using a Thioflavin T-based approach. New findings from a study comparing normal brain samples to those from patients with ALS reveal important insights into the disease. The study utilized the new HTRF Total Ataxin 2 and the Mutant Ataxin 2 Detection Kits to analyze protein samples. Results showed that the Ataxin 2 protein was more highly expressed in the ALS patient sample, with a high aggregation rate. Notably, the mutant form of the protein was only detectable in the ALS patient sample, and a disaggregation step resulted in a positive assay signal. These findings provide a better understanding of the role of Ataxin 2 in ALS and could lead to new approaches for treating the disease. In this paper, we explored the effects of a synthetic steroid, pregnanolone glutamate (PAG), and its metabolites on the brain and behavior. By identifying these metabolites in rats and investigating their impact on neurotransmitter receptors in neurons, along with studying their effects on the behavior of zebrafish, we observed that these compounds can influence receptors and impact behavior. 🧠Migraine is among the most common neurological disorders. Two independent neuropeptides have received significant attention for their role in migraine pathogenesis: calcitonin gene-related peptide (CGRP) and pituitary adenylate cyclase-activating polypeptide (PACAP).
