Due to its high affinity and stability, the streptavidin-biotin interaction is frequently leveraged to purify and detect biomolecules. However, its application with #labelfree #biosensors such as BLI and #SPR has been limited by the irreversibility of the interaction. IBA Lifesciences' Twin-Strep-tag® Capture Kit uses a modified streptavidin molecule to retain high affinity and stability while allowing reversibility with SPR regeneration conditions. In our last video, we showed how we can simultaneously merge thousands to tens-of-thousands of droplets by field-of-view, or across the entire array, to bring the contents together with high precision. We also showed how sequential merges can be performed to build multi-step workflows to meet a wide range of experimental needs. So, having brought the contents together, what types of readouts can we capture? The Lightcast platform utilizes brightfield and a selection of fluorescence filters to support a range of functional assays such as antigen binding or cytokine secretion for hit identification. Real-time processing enables sequential processing, down-selection (whereby only a sub-set of droplets move to the next step of the workflow), or dispense.
