T cells play a pivotal role in immune defense and cancer surveillance. A golden standard for enumeration of activated T cells is ELISpot. By defining the right assay parameters, ELISpot read-outs are highly informative. This golden standard method is cost-effective and ideal to support clinical studies. In our recent publication, Julian Freen-van Heeren, PhD; @Irma Rensink Annelies Turksma; Anja ten Brinke of our R&D team, share our insights for setting up an IFN-γ ELISpot that allows precise enumeration of antigen-specific T cells. As model system we assessed SARS-CoV-2-specific T cell responses and compared pre-pandemic and post-recovery T cell responses. A #translationalmedicine (preprint) study identifying CCL3/4 as biomarkers for delayed healing and CCR5 as a potential target for immunotherapies in cutaneous leishmaniasis. ▶ We analyzed clinical RNAseq of skin lesions and stratified patients according to the expression of chemokine genes CCL3/4, revealing that high expression delays the healing almost 2x! ▶ Back-translation with validation in mice showed that CCR5+ CD8+ cytotoxic cells are primarily recruited by those chemokines contributing to exaggerated inflammation 🔥 . ▶ Lesional IL-15 is crucial to up-regulate CCR5 expression in incoming CD8 T cells. ▶ Blockade of CCR5 experimentally with selective inhibitors improves healing dramatically! Current understanding of the immunopathogenesis and pathophysiology of anaphylaxis focuses on the roles of IgE and IgG antibodies, immune effector cells, and mediators thought to contribute to examples of the disorder. Human subjects including mice are antibodies genetically deficient in the antibodies, antibody receptors, effector cells, or mediators implicated in anaphylaxis, and mice that have been humanized for some of these elements.
