Historically, rocking motion bioreactors have been the preferred choice for culturing CAR-T cells due to their user-friendly nature and lower capital investment. However, with the emerging trend of large-scale production in allogeneic #cartcelltherapy, there is a need to explore more suitable alternatives. In a recent study, conducted in collaboration with University College London, the possibility of culturing CAR-Ts under stirred conditions was explored and an optimized protocol was developed. The results, presented in the webinar linked below, reveal that CAR-Ts cultured in the #Ambr®250 under stirred conditions demonstrated a higher fold expansion without affecting CAR-T expression, compared to those cultured in flasks. The study also compared pre-expanding the cells for 7 vs. 3 days prior to the stirred fed-batch culture. The IQue®3 flow cytometric analysis showed that CAR-T cells pre-expanded for 3 days exhibited a less differentiated phenotype (more naïve than effector) and lower expression of exhaustion markers (CD69, PD-1). The killing capacity was evaluated using the #Incucyte® live-cell imager, showing that the shortened protocol increased the production of TNF. To further increase the final yield, a process intensification method was studied using the #MODDE®, design of experiments tool. The shortened fed-batch protocol was compared with perfusion culture. Perfusion using #4Cell®Nutri-T XF GMP medium achieved 4.5X yields compared to fed-batch cultures after 7 days. Importantly, the CAR-Ts obtained by perfusion retained ideal therapeutic characteristics such as a less differentiated, less exhausted, and less activated phenotype. The next steps include scaling up the process to 2L in the #Univessel®STR Bioreactor and automating the harvesting step using #KSep®400.
