We need to further define the terms 'organoids' and 'spheroid' cells. Personally, I disagree with labeling patient-derived cells as 'organoids,' as discussed in this paper. We are excited to announce initial Phase I data of JBI-802 a CoREST inhibitor, which shows therapeutic potential in immunotherapy resistant NSCLC patients with poor prognosis based on their genetic and metastatic pattern. In addition, the on-target dose/exposure-proportional decrease in platelet constitute a proof-of-principle that JBI-802 can be an active compound in hematological malignancies like Essential Thrombocythemia (ET) and other MPN/ MDS characterized by thrombocytosis. A follow up Phase I/II study in MPN/ET and MPN/MDS with thrombocytosis is being planned in first quarter of this year to investigate JBI-802 as potential novel treatment option. The currently available #chimeric #antigen #receptor ( #CAR) #T #cell #therapies have had limited efficacy for a wide range of #solid #cancers and #hematological #malignancies, due in part to CAR T cell #dysfunction. The #scavenger #receptor CD5 is associated with the T cell receptor ( #TCR) and inhibits T cell function. Patel et al. show that #genetic_deletion of #CD5 in #CAR and #TCR T cells enhances expansion, persistence, and #cytotoxicity and is #effective in preclinical solid tumor and hematological malignancy models. For July's #DDU journal club we're highlighting a recent #jmedchem drug annotation describing the development and optimisation of the clinical candidate Sonrotoclax for B-cell malignancies. Sonrotoclax is a protein-protein inhibitor of Bcl-2, a key mediator of cellular apoptosis. It prevents Bcl-2 from sequestering proapoptotic proteins, promoting apoptosis of cancerous cells. The authors used co-crystal structures to design molecules with a novel binding mode in the P2 pocket, distinct to previously disclosed inhibitors, including the clinical approved Bcl-2 inhibitor Venetoclax. An elegant optimisation campaign lead to Sonrotoclax which is undergoing evaluation in Phase II/III clinical trials in several hematologic cancers. https://lnkd.in/eGDkW_Fj

This study of real-world metastatic nonsquamous NSCLC patients shows statistically significant longer overall survival in patients who have molecular genotyping results available before 1L therapy. This emphasizes the importance of early molecular diagnostic testing for targeted therapy selection. TIL therapy originated from research at the National Institutes of Health in the 1980s. It took decades to fine-tune, and Iovance had to overcome multiple regulatory delays bringing the therapy, formerly known as lifileucel, to market.
