💡 Did you know Immunofluorescence (IF) bleed-through can significantly hamper image scan quality? When fluorophores in IF studies have closely matched wavelengths, there's a risk of light from one channel inadvertently affecting another, a phenomenon known as 'spectral overlap'. This overlap complicates image analysis, making it challenging to pinpoint precise staining for IF markers. Such disruptions can hinder the overall progress of a study. Before beginning quantitative analysis, our image analysts strongly recommend to first conduct a thorough QC review of all images to assess suitability for analysis, and to only proceed with images that meet certain criteria. There seems to be disagreement on what is what,, but from what I've read he basal lamina and the basement membrane are not the same thing. The former is part of the latter, comprising the layer proximal to the cell membrane. The aim of this study was to test the expression of RKIP in different subtypes of leiomyoma (usual type, cellular, apoplectic or hemorrhagic leiomyoma, leiomyoma with bizarre nuclei and lipoleiomyoma) and leiomyosarcoma specimens and to determine its biological role in leiomyosarcoma cells. The team, led by Pasquapina Ciarmela conclude that the difference in RKIP expression in different cases and histological types underscores the importance of continuing to study RKIP as a possible marker candidate in myometrial histopathological studies. Their study is one of the first to investigate the biological role of RKIP in leiomyosarcoma, and it was seen that its downregulation seems to favor the clonogenicity in the leiomyosarcoma.
