Could a novel therapeutic targeting NMDAR hypofunction improve cognition in patients with neurodegenerative disorders? Congratulations to Jacob Beckley, Teresa Aman, Michael C. Quirk, and their team at Sage Therapeutics for the recently published paper disclosing a NMDAR positive allosteric modulator, SAGE-718, and supporting it as an efficacious therapy for the treatment of NMDAR hypofunction. Interesting therapeutic approach from UCLA for neurodegenerative diseases (dementia/Alzheimer's) that works differently from the recently FDA-approved antibodies (Lecanemab, Donanemab) which target amyloid beta plaques in the brain. This approach involves a therapeutic small molecule that enhances γ-oscillations (30 to 120 Hz) and improves cognition and memory in mice by blocking the GABA-A receptor. This receptor typically inhibits the interneurons that generate γ-oscillations. Looking forward to seeing future clinical trials, if any are planned.
