While fluorescent labeling of EVs is quite a preferred (and necessary) practice for many research areas including diagnostics and therapeutics, traditional way of labeling comes with certain limitations including non-specific binding, stability issues, and photobleaching of fluorophores. However at HansaBioMed Life Sciences Ltd, we provide EV researchers with naturally-fluorescent EVs, secreted from engineered cells. Thanks to this cell enineering, our FLUO-EVs express EGFP (green), BFP (blue), or mCherry (red) as fusion proteins with CD9, CD63, and CD81. As a result, these FLUO-EVs do not bleach, stay stable, and do not possess the risk of measuring non-specific binding. 🔬 Delving into the intricacies of therapeutic oligonucleotides (ONs), this study sheds light on the critical role of higher-order structures in the separation of ON diastereomers using HILIC. The research reveals how factors like column temperature and mobile-phase ionic strength can be fine-tuned to either enhance or suppress diastereomer separation, significantly advancing the characterization of PS-containing ONs. Expamers for T Cell Activation and Next Gen CAR-T Have you heard? Expamers are new drug substances being used to prepare a next generation of CAR-T. They are modular complexes that simultaneously "touch" and activate multiple cell receptors. 📔 Here's a paper by Mateusz Poltorak while he was a lead investigator at Bristol Myers Squibb. It was just shared with me the other day, and I was eager to check it out: https://lnkd.in/edCEQW_t - Expamers are ~100 nm complexes made of a Strep-Tactin backbone and anti-CD3/CD28 Ab fragments. - The backbone is polymeric and the Fab domains are tagged with a Strep-tag, allowing them to self-assemble. - They’re soluble and distribute evenly in cultures. Activation is stopped by disruption with biotin.
