This November, capivasertib (TRUQAP®) received approval by the FDA as a medicine for treating estrogen receptor (ER)-positive metastatic breast cancer (MBC) in adults. It acts as an inhibitor of AKT1, AKT2, and AKT3 isoforms of serine/threonine kinase AKT (protein kinase B) that results in inhibiting the phosphorylation of substrates in the AKT signaling pathway. The AKT kinase plays a crucial role in regulating many processes including metabolism, cell survival, growth, and proliferation. Mutations and loss of functions in the AKT pathway are among the most common reasons for breast cancer. Capivasertib contains piperidine-4-amine moiety in its backbone. Embryo diapause is a unique biological phenomenon as a survival strategy to enhance reproductive fitness in adverse environments. Recent studies indicate that tumor cells also exhibit a similar state when exposed to chemotherapy drugs, but the molecular mechanisms underlying the transition of tumor cells to a diapause-like state still require further investigation. Earlier this year, a collaborative study published in Cell Metabolism (IF 29.0) provided new insights into the non-hereditary mechanisms of chemotherapy resistance. Despite the success of delivery technologies such as lipid nanoparticles and trivalent GalNAc conjugation, targeted delivery of oligonucleotides to extra hepatic tissues remains a challenge. At this year’s OPT Congress, WuXi AppTec presented a poster showing the utilization of our peptide-based DNA-encoded library screening platform to identify endocytosis receptor-binding peptides. The binding affinity of hit peptides was confirmed through biophysical assays, and target-based delivery efficiency in breast cancer cells was evaluated by flow cytometry.
