These molecules reduce brain inflammation, which in turn improves insulin function. New study shows a role for cholesterol in pain perception When you stub your toe or bump your head, you know that rubbing the injury can lessen the ouch. But how? New research shows how physical pressure on cells can reduce pain signals, while excessive cholesterol clumps in cell membranes can interfere with that process. "Excess cholesterol is a feature in many diseases and disorders, including diabetes and diseases of aging," said Hansen, an associate professor of molecular medicine at The Wertheim UF Scripps Institute. "This could be one explanation for why we see more chronic pain in these groups." Dysfunction of mitochondria is a key factor in the development of disease, especially with ageing, with exercise being the only known intervention to counter this. However, with regards to this essential organelle, can we get too much of a good thing? Some recent studies have suggested that high volumes of high intensity interval training (HIIT) may impact mitochondrial function as evidenced through impaired glucose tolerance, oxidative damage and a reduction in mitochondrial oxidative function. It should be noted that these outcomes were reversed quickly with a reduction in volume and that the participants had not conducted a period of preparation for the HIIT protocol. So, whilst HIIT is an effective approach for many populations, including clinical ones, proper progression is needed to prepare for it and active recovery, lower intensity days, need to be considered. The findings suggest that resistance training may have potential benefits for individuals undergoing inelastic compression therapy for chronic venous disease, as it can improve the effectiveness of compression therapy by enhancing interface pressure and reducing venous hypertension during ambulation. Bridging the Gap Between Science and Practice: The Project BIG #Biorepository Blood, CSF, and stool were longitudinally collected from participants, prioritizing times of change - including relapsing or remitted state or change in disease modifying immunotherapy. PROs on pain, QOL, fatigue, anxiety/depression, and sleepiness were collected. Biospecimens and data were shared across the bedside-to-bench spectrum with bench scientists. The biorepository has 491 participants and is composed of 5 disease groups: #MultipleSclerosis (64%), NMO (4%), MOGAD (7%), Transverse Myelitis (2%), and Autoimmune Encephalitis (2%). MS participants had subthreshold or no depression, mild anxiety, moderate QOL, moderate fatigue, and moderate pain. There was a significant difference in pain scores, with MS participants scoring lower than the other groups. Chronic stress and diabetes mellitus are well-known for their roles in inducing oxidative stress and inflammation, leading to disruptions in cell membranes and platelet activity. Our findings provide valuable insights into how chronic psychological stress impacts platelet phospholipid membrane composition and platelet activation in type 1 diabetes. This contributes significantly to our understanding of the intricate interplay between stress, lipid metabolism, and diabetes management, showcasing the collaborative efforts of scientists from the #InnovativeMedicineCentre and #VilniusUniversity.
