June is Alzheimer’s & Brain Awareness Month. Alzheimer's disease and related dementias (AD/ADRD) pose a significant public health challenge, with devastating effects on individuals, families and communities. In 2020, as many as 5.8 million individuals aged 65 or older in the US were living with AD/ADRD. By 2050, this number is projected to rise to 13.8 million. A comprehensive study is currently underway to clarify the relationship between Alzheimer’s disease and related dementias (AD/ADRD) and four toxic elements: lead, mercury, arsenic, and cadmium. These elements, often encountered by humans and animals worldwide without their knowledge, may hold crucial insights into cognitive decline. The study, led by Aaron Specht, Assistant Professor in the Purdue University School of Health Sciences, aims to uncover the intricate connections between exposure to these toxic substances and the progression of cognitive impairment over time. This new study is one of the largest, most representative, and most racially diverse studies investigating the impact of toxic metal exposure on AD/ADRD in humans to date. Traditionally, Alzheimer’s disease has been linked to the accumulation of beta-amyloid in the brain, with the buildup of the protein seen as a hallmark of the disease. However, Baroness Susan Greenfield CBE, NeuRA’s expert in residence this week, has co-authored a paper, ‘Evidence for a novel neuronal mechanism driving Alzheimer's disease, upstream of amyloid’ published in Alzheimer's & Dementia: The Journal of the Alzheimer's Association®, which explores the possibility of even earlier trigger, which may lead to treatments that prevent the build-up of beta-amyloid from starting. The paper reviews evidence for this upstream mechanism, including the role of a little-known neurochemical in driving the process.
