An Unexpected Membrane Phase Transition It is always very exciting when a new physical phenomenon is discovered in lipid membranes, especially when the phenomenon cannot yet be explained. John Straub, Björn M. Reinhard, and colleagues report on cholesteryl alkynes that change the phase behavior of membranes: the membranes are disordered at low temperatures and in an ordered state at higher temperatures, the opposite of what we normally see. It is a combination of many effects: a dependence on the ensemble of lipids surrounding the cholesteryl alkynes, the orientation of the alkynes, the saturation of the phospholipids, the polarity of water (which decreases with higher temperatures), and many unknowns yet. The paper triggered two ideas in my head: - Can we exploit the effect seen here in drug delivery? Can we design molecules that flip-flop in the membrane depending on a trigger and thus change the surface characteristics of LNPs or liposomes? - How do local temperature fluctuations in membranes and proteins influence the polarity of water?
