A masterpiece in enzymology, what a pleasure to see this in the publication landscape!
👀 😀 Always gratifying to witness the impact of the crucial role of #bindingkinetics in efficacy prediction! In this impresive publication, researchers provide mechanistic insights for the largest cohort of EZH2 inhibitors reported to date, demonstrating that 𝙧𝙚𝙨𝙞𝙙𝙚𝙣𝙘𝙚 𝙩𝙞𝙢𝙚 values obtained from biochemical assays are the 𝗯𝗲𝘀𝘁 𝗽𝗿𝗲𝗱𝗶𝗰𝘁𝗼𝗿 𝗼𝗳 𝗰𝗲𝗹𝗹𝘂𝗹𝗮𝗿 𝗲𝗳𝗳𝗶𝗰𝗮𝗰𝘆. AUC values obtained from a cellular washout assay together with endpoint IC50’s from the same assay were also good predictors for antiproliferative activity. https://lnkd.in/ddaKgxgU #DrugDiscovery #Screening #HTS #MedChem #Drugdesign #epigenetics

A new #preprint describing a thermostable RNase inhibitor: "Over the past decade, there have been remarkable technical improvements in most aspects of single-cell transcriptomics. Yet, there has been little to no progress in advancing RNase inhibition despite that maintained RNA integrity is critical during cell collection, storage, and cDNA library generation. Here, we demonstrate that a synthetic thermostable RNase inhibitor yields single-cell libraries of equal or superior quality compared to ubiquitously used protein-based recombinant RNase inhibitors" https://lnkd.in/dFq8hUva

PMOs, short for phosphorodiamidate morpholino oligomers, are short single-stranded DNA analogs that are built upon a backbone of morpholine rings connected by phosphorodiamidate linkages. They bind to complementary sequences of target mRNA to block protein translation through steric blockade. Their interference of viral protein translation operates independently of RNAse H and are resistant to a variety of enzymes present in biologic fluids, making them highly suitable for in vivo applications. PMO-based therapy for Duchenne muscular dystrophy (DMD) has become a hallmark for PMO-based antisense therapy. Here's a link to a nice write-up on the history on PMO's: https://lnkd.in/gvzUr6jv. as well as the challenges and strategies of Antisense Oligonucleotide Drug Delivery: https://lnkd.in/gQd2V5Bk
