I would have never suspected agar, if I didn't find colonies on my plate. (But this is how great discoveries are made!!!) Looks like bad agar batch(es) have been killing yeast in labs world over. And it dates back to 2006 (at UMass Chan Medical School) The culprit is probably an antifungal metabolite/compound in algae from which agar components are produced. There are already some clues... 1) Washing agar solves the problem - the compound is water soluble 2) Survives autoclaving, 121 C - unlikely to be a protein 3) The inhibitory effect is attenuated in rich media - maybe a competitive mechanism Researchers have even uploaded a paper on biorxiv with guidelines on troubleshooting and quality control. https://lnkd.in/eH8zPq9A
