I just completed Thermo Fisher Scientific's Genetic Sciences on Forage. I learned to analyze Real-time PCR tests and assess the sensitivity and specificity of a hypothetical disease. These job simulations are great and useful, and they are helping me develop the ability to avoid being overwhelmed by new information. In a real-world job, while the company would provide us with the necessary training, dealing with a new problem requires constant self-learning and patience to solve it. I have completed another Job Simulation by Thermo Fisher Scientific, this time in Genetic Sciences. I have broadened my knowledge into the process of PCR, whilst developing a holistic understanding of identifying critical markers used to analyse PCR data. This experience has allowed me to: - Complete a job simulation involving analysis of Real-time PCR tests for Thermo Fisher.  - Analyse amplification curves and determine cycle thresholds for different genes.  - Assess the sensitivity and specificity of a hypothetical disease. - Research Real-time PCR assays and analyse each test to understand their limits and positive percent agreements.

The hot springs in Yellowstone were a bit of a "pilgrimage" for me!!! As a molecular biologist undergraduate in the 1980s, I remember being terribly excited to learn about the PCR reaction for the first time!! PCR was made possible using the Taq polymerase enzyme from the thermophilic bacterium Thermus aquaticus which was first isolated by Thomas Brock from a hot spring very close to where I am in this picture, where it thrives at over 70 degrees Celsius! Sanger DNA sequencing which was used for the first human genome back in 2001 also uses Taq. And Taq polymerase became the basis for much of the Biotech industry we know today. If you had a Rapid PCR test during the COVID pandemic, Taq was used in that test and in many other diagnostic tests now used routinely. I feel I owe much of my career to this amazing discovery. Now that I work in clinical research, I see many of the amazing innovations which Taq enabled reaching the clinic.
