What's unique about Canagliflozin is that it isn't a typical senolytic that indiscriminately blasts away senescent cells, sometimes leading to side effects It's not even a senomodulator/senomorphic that basically "defangs" senescent cells by inhibiting secretion of pro-inflammatory and other troublesome factors (known as the SASP). But these don't actually remove the pathological senescent cells, which can later on re-activate secretion of SASP Canag acts as a Senescence-associated Immunomodulator -- which basically restores our immune system so it's better able to "naturally" target and clear out senescent cells. This is perhaps the most promising class of senotherapeutics because our immune system is very robust at regulating senescent cell burden when we're younger and becomes compromised as we age -- leading to increased senescent cell burden and age-related dysfunction. Canag basically "reminds" our immune system how to effectively clear senescent cells once.more

A study by the Buck Institute for Aging Research found that the accumulation of senescent cells contributes to aging and related diseases, but eliminating these cells can improve health. Research shows that inhibiting sodium-glucose co-transporter 2 (SGLT2) with canagliflozin enhances the clearance of senescent cells, improving age-related changes. In obese mice, canagliflozin reduced senescent cells in visceral fat, decreased inflammation, and improved metabolism, unlike insulin. It also extended the lifespan of prematurely aging mice. Canagliflozin upregulated a compound that boosted immune clearance of senescent cells by downregulating programmed cell death-ligand 1, suggesting that SGLT2 inhibition indirectly promotes the removal of senescent cells. #SenescentCells #Canagliflozin #SGLT2Inhibitor #LongevityScience #MetabolicHealth #ImmuneBoost #ObesityResearch
