🔬 Troubleshooting Hematoxylin & Eosin (H&E) Staining: Common Issues and Solutions 🔬 As histology professionals, we know that achieving optimal H&E staining is crucial for accurate tissue diagnosis. However, various issues can arise during the staining process. Here are some common issues and their solutions: ● Uneven Staining: ▪︎ Problem: Uneven staining can result from inconsistent section thickness or improper deparaffinization. ▪︎ Solution: Ensure uniform sectioning and thorough deparaffinization. Regularly check and maintain microtome blades and water baths. ● Weak Hematoxylin Staining: ▪︎ Problem: Weak nuclear staining may occur due to over-oxidized hematoxylin or insufficient staining time. ▪︎ Solution: Use fresh hematoxylin and monitor staining time closely. Adjust the pH of the staining solution if necessary. ● Overstained Eosin: ▪︎ Problem: Overstaining with eosin can obscure cellular details. ▪︎ Solution: Optimize eosin staining time and use appropriate differentiation steps. Regularly change eosin solutions to maintain consistency. ● Chatter and Exploding Sections: ▪︎ Problem: Chatter and exploding sections often result from over-processed or poorly infiltrated tissues. ▪︎ Solution: Adjust tissue processing protocols to ensure proper infiltration and avoid over-dehydration. ● Water Contamination: ▪︎ Problem: Water in alcohols or xylenes can cause lightening of eosin and pink haze artifacts. ▪︎ Solution: Regularly change reagents and monitor ambient humidity. Use high-quality, consistent water sources. By addressing these common issues, we can improve the quality and reliability of our H&E stains, ensuring better diagnostic outcomes.
