I clearly remember that in 2018, I talked with Diane McCarthy of the United States Pharmacopeia about AAV standard reference materials. At that time, it was very difficult to find good reference materials. Seven years later, AAV-related technology has been significantly improved. Under optimal transfection conditions, the yield of HEK293F and VPC cells can easily reach 10E10 vg/mL. I am pleased to see improvements in the quality of standards such as PROGEN which provides fully characterized AAV standards. The same standard materials were also used for method evaluation. Silja Syrbe's poster at ASGCT 2024 shows that reliable standards and their use in orthogonal methods (ELISA, BLI, SEC-MALS, DLS, CE) and throughout ensure not only the most precise results, but also the final The safety of the product complies with current regulatory requirements. Capsid titer with BLI is pleasant compared to other techniques and can be performed in a broader set of matrices. All you need is a good standard. That is available. If titer for AAV, both capsid AND genome, is a bother—call Gator.
