This is big news in the tissue processing community… check out this recent article about Xylene vs Isopropanol for Paraffin Wax Processing of Lung Tissue. It concludes that isopropanol, which is what we use in our instruments instead of xylene, can be used just as effectively as traditional xylene processing. The more labs who adapt this safer processing methods, the fewer people are exposed to xylene’s harmful effects. One of the reasons for instability in #lyophilisedformulations on storage and possibly increased reconstitution times is the change in the physical form of the crystalline excipient. Stabilising the crystalline component requires careful cycle design. 3D X-ray micro-CT is an ideal techniques to visualize the interior of drug products and assess CQAs important for Q3 bioequivalence. This micro-CT dataset reveals the complexities of a generic extended release capsule bead product, Venlafaxine (brand name Effexor). The coating on the beads, seen as a white layer in the scan below, is key to product performance. Taken straight from the drug sheet "Drug release is controlled by diffusion through the coating membrane on the spheroids." micro-CT analysis allows us to evaluate the quality, coverage, and consistency of the coating. In the scan below, we find instances of incomplete coating, fractured beads, and even internal void spaces. Visualizing and quantifying these features closely is key to assuring a succesful ANDA and product launch. Not doing so will lead to costly pitfalls in performance, product quality, and failed ANDA filings.
