📢[New Publication! – Optimizing #AAVPurification using the High-Throughput and Automated Crossflow System] 🧬Adeno-associated viral (AAV) vectors have become well-established tools in #genetherapy, driving the demand for faster and more efficient #AAV manufacturing. One critical step in #AAVmanufacturing is the ultrafiltration and/or diafiltration (UF/DF) during #downstreamprocessing, which typically requires substantial amounts of feeding material (i.e. AAV) and time for effective optimization. ⌛ To address this, researchers from iBET - Instituto de Biologia Experimental e Tecnológica and Sartorius, used the Ambr® Crossflow system for high-throughput, automated screening of multiple UF/DF parameters in one single run. Key highlights: ➡️Different membrane materials (PES and Hydrosart), pore sizes (30 and 100 kDa) and process conditions were investigated. ✅Best performer: 100kDa PES flat sheet cassette achieved the best overall performance. ✅Scalable performance: This cassette demonstrated similar AAV8 recovery, permeate flux and total protein removal to small-scale devices, and outperformed the hollow fibre option when scaled to a larger system. 💪The Ambr® Crossflow allowed to efficiently test various UF/DF parameters, making it a powerful accelerator for developing efficient AAV downstream processing.
